目的:比较不同剂量鱼藤酮损伤大鼠与脾气虚证大鼠海马组织c AMP/PKA信号通路的变化。方法:30只SPF级雄性大鼠,随机分为正常组,脾气虚组,鱼藤酮高、中、低剂量组(2.0、1.5、1.0m L/kg),每组6只。HE法观察各组大鼠海马形态变化,ELISA法检测各组大鼠海马组织c AMP含量,Real-time PCR法和Western Blot法检测蛋白激酶(PKA)、糖原磷酸化酶(Gp)、磷酸化酶激酶(PHK)的m RNA及其表达量。结果:与正常组大鼠比较,鱼藤酮各剂量组大鼠与脾气虚证组大鼠的海马组织形态变化不明显,其c AMP含量及PKA、Gp、PHK m RNA和蛋白的表达均有不同程度的减弱,3组中鱼藤酮中剂量组的结果与脾气虚证组具有一定相似性。结论:中剂量鱼藤酮损伤大鼠海马c AMP/PKA信号通路的变化与脾气虚证大鼠之间存在关联性,脾气虚证大鼠海马c AMP/PKA信号通路的改变可能与调控代谢相关基因Gp、PKA、PHK的表达有关。
OBJECTIVE: To compare the changes of c AMP / PKA signal pathway in hippocampus of rotenone rats with different doses of rotenone and rats with spleen-qi deficiency syndrome. Methods: Thirty SPF male rats were randomly divided into normal group, spleen deficiency group, high, medium and low dose rotenone groups (2.0,1.5,1.0 m L / kg), with 6 rats in each group. The hippocampal morphological changes of rats in each group were observed by HE method. The content of cAMP in hippocampus of rats in each group was measured by ELISA. The protein kinase (PKA), glycogen phosphorylase (Gp) The m RNA of the enzyme kinase (PHK) and its expression level. Results: Compared with normal rats, the morphological changes of hippocampus were not obvious in rats of rotenone group and those of spleen-qi deficiency group, and the expression of cAMP, PKA, Gp, PHK m RNA and protein were all different Of the three groups of rotenone dose group results and spleen qi deficiency group has a certain similarity. Conclusion: The changes of c AMP / PKA signal pathway in rat hippocampus induced by medium dose rotenone have a relationship with the rats with spleen deficiency syndrome. The changes of cAMP / PKA signal pathway in hippocampus of rats with spleen deficiency syndrome may be related to the regulation of metabolism related gene Gp , PKA, PHK expression.
