目的:研究兔子宫内膜是否存在干细胞。方法:流式细胞术检测兔子宫内膜中上皮细胞及间质细胞的特异性标记分子,干细胞培养基培养出富含干细胞的集落,运用RT-PCR方法检测分化前后相关分子的表达,MTT法检测集落细胞分化前后增殖能力变化,流式细胞术检测分化前后的相关分子表达变化及细胞的凋亡情况。结果:兔子宫内膜存在上皮细胞及间质细胞两种细胞,干细胞培养基可培养出富含干细胞的集落,且分化前的细胞具有更高的增殖潜能,表现出更多的干细胞源性,具有更低的细胞凋亡比例。结论:兔子宫内膜存在富含干细胞的集落,提示存在子宫内膜干细胞。
Objective: To study the existence of stem cells in rabbit endometrium. Methods: Flow cytometry was used to detect the specific markers of epithelial cells and stromal cells in rabbit endometrium. Stem cell-rich colonies were cultured in stem cell medium. The expression of related molecules before and after differentiation was detected by RT-PCR. MTT assay The proliferation of colonies were detected before and after differentiation. Flow cytometry was used to detect the changes of related molecules before and after differentiation and the apoptosis of cells. Results: There were two types of epithelial cells and stromal cells in rabbit endometrium. Stem cell culture medium could grow stem cells rich in stem cells, and the cells before differentiation had higher proliferative potential and showed more stem cell origin, Has a lower proportion of apoptotic cells. Conclusion: There are stem cell-rich colonies in rabbit endometrium, suggesting the presence of endometrial stem cells.
