本实验采用脾集落形成法和流式细胞术等技术,研究了小鼠颌下腺组织培养上清液(SGCM)对造血干细胞和骨髓细胞增殖的影响。结果表明:正常雄性和雌性SGCM对小鼠造血干细胞增殖和分化有明显促进作用;SGCM能有效促进小鼠骨髓细胞(BMC)由相对静止期(G0/G1)进入增殖活跃期(S+G2/M),并能提高骨髓有核细胞数。研究提示,正常小鼠颌下腺可能通过分泌造血生长因子样物质促进小鼠造血干细胞增殖和分化并加快BMC从静止期进入增殖周期,从而发挥造血调控功能。
In this study, spleen colony formation and flow cytometry and other techniques to study the mouse submandibular gland tissue culture supernatant (SGCM) on the proliferation of hematopoietic stem cells and bone marrow cells. The results showed that normal male and female SGCM could significantly promote the proliferation and differentiation of hematopoietic stem cells in mice. SGCM could effectively promote the migration of mouse bone marrow cells (BMC) from the quiescent phase (G0 / G1) to the proliferative phase (S + G2 / M) , And can improve the number of bone marrow nucleated cells. Research suggests that the normal submandibular gland may exert its hematopoietic regulation function by promoting the proliferation and differentiation of hematopoietic stem cells and accelerating BMC from quiescent to proliferative cycle by secreting hematopoietic growth factor-like substances.
