目的探讨JAZF1过表达对鼠肝细胞IAR-20及Hepa1-6糖脂代谢相关基因的影响。方法克隆鼠JAZF1编码区并构建pIRES2-EGFP-JAZF1表达载体,利用脂质体法将目的基因转染到鼠肝细胞IAR20及Hepa1-6中,采用实时荧光定量PCR测定各处理组JAZF1 mRNA表达情况,以及糖脂代谢相关基因SREBP1、ACC、FAS、HSL、PPARα、ATGL、GluT-1、GluT-4和细胞因子FGF 21mRNA的变化情况。结果 JAZF1基因在鼠肝细胞中过表达导致脂代谢基因SREBP1,ACC,FAS表达降低(P<0.01),PPARα和HSL表达增加(P<0.01),对ATGL无影响(P>0.05),使糖转运相关基因GluT-1表达增加(P<0.01),GluT-4无显著变化(P>0.05),对FGF-21无影响(P>0.05)。结论 JAZF1过表达可以抑制肝细胞的脂肪生成,促进脂肪分解,并可能增加基础葡萄糖转运。
Objective To investigate the effect of JAZF1 overexpression on the related gene of glucose metabolism in rat hepatocytes IAR-20 and Hepa1-6. Methods The coding region of JAZF1 was cloned and the pIRES2-EGFP-JAZF1 expression vector was constructed. The target gene was transfected into rat hepatocytes IAR20 and Hepa1-6 by lipofectamine. The expression of JAZF1 mRNA in each treatment group was detected by real-time fluorescence quantitative PCR , And the changes of glucose and lipid metabolism related genes SREBP1, ACC, FAS, HSL, PPARα, ATGL, GluT-1, GluT-4 and cytokine FGF21 mRNA. Results Overexpression of JAZF1 in rat hepatocytes resulted in a decrease of SREBP1, ACC and FAS expression (P <0.01), increased expression of PPARα and HSL (P <0.01) and no effect on ATGL (P> 0.05) GluT-1 expression increased (P <0.01), GluT-4 had no significant change (P> 0.05), but had no effect on FGF-21 (P> 0.05). Conclusion JAZF1 overexpression can inhibit lipogenesis in hepatocytes, promote lipolysis, and may increase basal glucose transport.
