BACKGROUND: There is increasing evidence that CD44 can be used as a specific marker of colorectal adenocarcinoma stem cells. In recent years, it has been found that embryonic stem cell transcription factor Oct4 is expressed in colorectal adenocarcinoma. Objective: To observe the number, location and distribution of CD44 + / Oct4 + cells in primary colorectal adenocarcinoma. Methods: 108 specimens of colorectal adenocarcinoma, 18 specimens of adjacent normal mucosa and 18 specimens of colorectal adenoma with atypical hyperplasia were made into three groups at 48 points. Immunohistochemical double staining and hematoxylin-eosin staining The eosin staining and localization of CD44 + / Oct4 + tumor cells were observed and their morphological characteristics were observed at the corresponding locations of hematoxylin-eosin chips. RESULTS AND CONCLUSION: Immunohistochemical double staining showed that there was no CD44 + / Oct4 + cells in adjacent normal mucosa adjacent to the tumor. A very small number of CD44 + / Oct4 + cells were observed in adenomas with atypical hyperplasia. CD44 + / Oct4 + cells. Double positive cells were mainly distributed in the side of the basement membrane of adenoid-like structure or in the co-wall side of the co-wall glands, with small or scattered dot-like distribution. The cells were oval or cuboidal with few cytoplasm and nuclei Deep stained, oval or tall columnar. The number was negatively correlated with the degree of colorectal cancer differentiation (r = -0.579, P <0.01), which correlated with the depth of tumor invasion (r = 0.236, P <0.05). The results suggest that CD44 + / Oct4 + cells may be colorectal adenocarcinoma stem cells.
