目的初步了解2014年山西省运城地区病毒性脑炎患者中乙型脑炎病毒E基因分子生物学特征和基因型别分布特征。方法采集19例临床诊断为病毒性脑炎患者的脑脊液标本,提取核酸通过巢式聚合酶链式反应(n PCR)扩增乙型脑炎病毒基因片断,采用生物信息学软件对病毒E基因区段核苷酸和氨基酸序列进行分子生物学特征分析并与通过构建系统进化树与国内外不同年代和地域分离的覆盖5个基因型别的乙型脑炎病毒进行基因型别研究。结果 19例山西运城脑炎患者标本中有2例为乙型脑炎病毒核酸阳性,基因分型结果显示均为基因Ⅰ型乙型脑炎病毒。结论 2014年山西运城地区病毒性脑炎患者中乙型脑炎病毒为基因Ⅰ型,与毒力相关的8个位点与乙型脑炎病毒P3一致,没有发生突变,不会影响病毒毒力。理论上表明,目前使用的乙型脑炎病毒疫苗对当地的乙型脑炎病毒仍具有保护力。
Objective To understand the molecular biological characteristics and genotypic distribution of Japanese encephalitis virus E in patients with viral encephalitis in Yuncheng, Shanxi province in 2014. Methods Nineteen CSF samples were collected from patients with viral encephalitis. Nucleic acids were extracted from the encephalitis virus by nested polymerase chain reaction (n PCR). Bioinformatics software was used to analyze the E gene region The nucleotide and amino acid sequences were analyzed by molecular biology and genotyped with 5 Japanese encephalitis viruses isolated from different generations and regions at home and abroad by constructing phylogenetic tree. Results 19 cases of Shanxi Yuncheng encephalitis patients in 2 cases of positive for Japanese encephalitis virus nucleic acid, genotyping results showed that all genotype I Japanese encephalitis virus. Conclusions In 2014, patients with viral encephalitis in Yuncheng area of Shanxi province were genotype Ⅰ genotypes, and virulence-related 8 sites were identical with that of Japanese encephalitis virus P3, with no mutation and no influence on viral virulence . Theoretically, the currently used Japanese encephalitis virus vaccine still has protection against the local Japanese encephalitis virus.
