目的检测病毒性心肌炎小鼠不同时间点血清中半乳糖凝集素-9(Galectin-9)的水平及心肌组织中Galec-tin-9mRNA的表达,探讨Galectin-9在病毒性心肌炎发生发展中的作用。方法选取58只5~6周BALB/c小鼠,随机分为病毒组50只和对照组8只。病毒组小鼠腹腔接种0.10mL TC ID50为10-5/L的柯萨奇病毒B3(CVB3)N ancy株病毒液,分别于接种病毒后第7、10、14、28天处死8只小鼠;对照组小鼠腹腔接种0.10mL不含病毒的Eag le氏液,28d时处死小鼠。取心脏,在光镜下观察心肌病理变化,计算心肌组织病理学积分。取外周血,用ELISA检测血清中Galectin-9的含量。采用实时荧光定量PCR(RQ-PCR)检测心肌组织中Galectin-9mRNA的表达。结果病毒组小鼠血清中Galectin-9含量在第7、10、14天均较对照组明显增高(P<0.01)。病毒组小鼠心肌组织Galectin-9mRNA的表达在第7、10天均较对照组增强(P<0.01)。病毒组小鼠血清中Galectin-9的含量及心肌组织中Galectin-9mRNA的表达均与心肌组织病理积分呈正相关(r分别为0.75,0.74;P<0.05)。病毒组小鼠血清中Galectin-9的含量与心肌组织中Galectin-9mRNA的表达呈正相关(r=0.71,P<0.05)。结论急性期病毒性心肌炎小鼠Galectin-9表达明显升高,且与心肌炎病变严重程度呈正相关,Galectin-9可能在病毒性心肌炎的发生发展中起一定作用。
Objective To detect the level of galectin-9 in serum and the expression of Galec-tin-9 mRNA in myocardium of mice with viral myocarditis at different time points and to explore the role of Galectin-9 in the development of viral myocarditis . Methods Fifty-five BALB / c mice aged 5-6 weeks were randomly divided into two groups: 50 in the virus group and 8 in the control group. The mice in the virus group were inoculated intraperitoneally with 0.10 mL Coxsackievirus B3 (CVB3) N ancy virus solution with a TC ID50 of 10-5 / L, and 8 mice were sacrificed on days 7, 10, 14 and 28 after inoculation with the virus The mice in control group were inoculated intraperitoneally with 0.10 mL Eagle’s solution without virus and mice were sacrificed 28 days later. Take the heart, observe the pathological changes of myocardium under light microscope and calculate the histopathological score of myocardium. Peripheral blood was taken and serum levels of Galectin-9 were measured by ELISA. Real-time quantitative PCR (RQ-PCR) was used to detect the expression of Galectin-9mRNA in myocardium. Results The level of Galectin-9 in the virus group was significantly higher than that in the control group on the 7th, 10th, and 14th days (P <0.01). The expression of Galectin-9mRNA in the myocardium of the virus group was higher than that of the control group on the 7th and 10th days (P <0.01). The levels of Galectin-9 in serum and the expression of Galectin-9mRNA in myocardium were positively correlated with myocardial histopathological scores (r = 0.75 and 0.74, P <0.05, respectively). The level of Galectin-9 in serum of the virus group was positively correlated with the expression of Galectin-9mRNA in myocardium (r = 0.71, P <0.05). Conclusion The expression of Galectin-9 in acute viral myocarditis mice was significantly increased, and it was positively correlated with the severity of myocarditis. Galectin-9 may play a role in the development of viral myocarditis.
