目的探讨DEX(地塞米松)对缺氧缺血性脑损伤新生鼠脑组织AQP-4表达的影响,为临床治疗新生儿缺氧缺血脑病提供实验和理论依据,揭示其发病机制,以期指导该病的早期治疗。方法将健康7日龄SD新生大鼠96只,清洁级,雌雄不限,平均体重12-18g,随机分为假手术组,HIBD组,DEX组,各组32只,分离左侧颈总动脉并结扎,吸入92%N2+8%O2氮氧混合气体行缺氧处理3h,制成HIBD模型。假手术组只作分离左侧颈总动脉而不结扎,不作缺氧处理,不给与药物处理。DEX治疗组于建模成功后立即腹腔注射10mg/kg,假手术组及HIBD组大鼠腹腔注射等量生理盐水,上述各组均于术后6h、24h、48h、72h随机抽取8只,于麻醉下处死取脑组织,通过Western blot方法检测新生大鼠脑组织AQP-4蛋白表达及实时定量PCR检测AQP-4 mRNA的表达。结果 SD鼠在缺氧处理后出现异常行为学改变。AQP-4 mRNA及蛋白的表达在缺氧诱导后显著升高,DEX的治疗则使SD鼠异常行为的表现程度减轻,后趋于正常,且能够降低HIBD导致的AQP-4的升高。结论 DEX对HIBD的治疗具有一定的疗效。
Objective To investigate the effect of dexamethasone on the expression of AQP-4 in brain tissue of neonatal rats with hypoxic-ischemic brain damage and to provide experimental and theoretical basis for clinical treatment of neonatal hypoxic-ischemic encephalopathy and to reveal its pathogenesis and guidance Early treatment of the disease. Methods Ninety-six SD neonatal rats of 7 days old were randomly divided into sham operation group, HIBD group and DEX group, with a mean of 12-18g. And ligation, inhalation of 92% N2 + 8% O2 nitrogen and oxygen mixed gas hypoxia treatment 3h, made HIBD model. Sham operation group only for the left common carotid artery without ligation, not for hypoxia, not given drug treatment. The rats in DEX treatment group were given intraperitoneal injection of 10mg / kg intraperitoneally immediately after modeling. Rats in sham operation group and HIBD group were given intraperitoneal injection of normal saline. All the above groups were randomly selected at 6h, 24h, 48h, 72h after operation. The brain tissues were sacrificed under anesthesia. The expression of AQP-4 protein in brain tissue of neonatal rats was detected by Western blot and the expression of AQP-4 mRNA was detected by real-time quantitative PCR. Results Abnormal behavioral changes occurred in SD rats after hypoxia. The expression of AQP-4 mRNA and protein was significantly increased after hypoxia induction. The treatment with DEX lessened the abnormal behavior of SD rats and became normal afterwards, and decreased the HIBD-induced AQP-4 elevation. Conclusion DEX has a certain effect on the treatment of HIBD.
