目的 了解家兔颅脑创伤后内皮素及其A受体在脑血流量变化中的作用及意义。方法 家兔致以左额顶叶为着力部位的加速性撞击伤,动态观察脑组织内皮素含量变化、内皮素A受体mRNA表达和定位变化,内皮素A受体拮抗剂BQ123的作用,及上述变化对脑血流量变化的影响。结果 脑创伤后局部脑组织内皮素含量显著升高;内皮素A受体mRNA主要分布于脑血管内皮细胞,表达显著增强;创伤后局部脑血流量显著降低,应用BQ123治疗的动物,脑血流量下降被明显抑制。结论 内皮素通过其A受体的调节在颅脑创伤后脑缺血的形成中具有重要作用;应用内皮素A受体拮抗剂可防治颅脑创伤后脑缺血。
Objective To investigate the role and significance of endothelin and its receptor A in the changes of cerebral blood flow after traumatic brain injury in rabbits. Methods The rabbits were subjected to accelerated impact injury on the left frontal parietal lobe. Dynamic observation of endothelin content, endothelin A receptor mRNA expression and localization, and endothelin A receptor antagonist BQ123 in rabbits were performed. The above changes affect the changes of cerebral blood flow. Results After traumatic brain injury, the content of endothelin in local brain tissue increased significantly. Endothelin A receptor mRNA mainly distributed in cerebrovascular endothelium and the expression was significantly increased. The local cerebral blood flow decreased significantly after trauma. The blood flow of animals treated with BQ123 Decline was significantly inhibited. Conclusion The regulation of endothelin through its A receptor plays an important role in the formation of cerebral ischemia after traumatic brain injury. Endothelin A receptor antagonist can prevent cerebral ischemia after traumatic brain injury.
