应用间接免疫荧光法和显微荧光分光光度技术,研究了试管内无细胞毒性浓度的亚硒酸纳(硒)对来源于伯基特淋巴瘤的Raji细胞株,EpsteinBar病毒(EBV)抗原表达的影响以及抑制EBV转化人脐血B淋巴细胞(BL)的作用。结果表明,001~01mg/L的硒使Raji细胞的EA抗原抑制353%~501%;001~05mg/L的硒令EBNAMFS值降低84%~218%;01~10%mg/L的硒明显抑制EBV转化BL,抑制率为633%~921%。由于EBV与鼻咽癌(NPC)密切相关,NPC病人血清和头发中硒含量明显低于健康人。本研究提示:对NPC患者和EBV基因表达活跃的人群适当补硒,可能有助于NPC和与EBV相关疾病的预防和辅助治疗。
Indirect immunofluorescence and microscopic fluorescence spectrophotometry were used to study the in vitro cytotoxic concentration of selenite (selenium) against Raji cell line derived from Burkitt’s lymphoma and EpsteinBar virus (EBV) antigen. The effects of expression and inhibition of EBV transformed human umbilical cord blood B lymphocytes (BL). The results showed that 001~01mg/L selenium inhibited the EA antigen in Raji cells by 353%~501%; 001~05mg/L selenium reduced the EBNAMFS value by 8. 4%~218%; 01~10%mg/L selenium significantly inhibited the conversion of EBV to BL, and the inhibition rate was 633%-921%. Because EBV is closely related to nasopharyngeal carcinoma (NPC), the selenium content in serum and hair of NPC patients is significantly lower than that of healthy individuals. This study suggests that appropriate supplementation of selenium in NPC patients and those with active EBV gene expression may contribute to the prevention and adjuvant treatment of NPC and EBV-related diseases.
