目的:探讨脑脉舒康胶囊对短暂性脑缺血发作小鼠模型的影响。方法:采用昆明种小鼠,分为脑脉舒康胶囊组(1.8,0.9,0.45 g.kg-1)、模型组、假手术和阳性药(血栓通1.125 g.kg-1、尼莫地平0.03 g.kg-1)对照组,各组连续灌胃7 d,于第8天给药1 h后,结扎两侧颈总动脉30 min,并进行再灌注15 min制备短暂性脑缺血模型;眼球取血,测定超氧化物歧化酶(SOD)活性、丙二醛(MDA)含量;取部分小鼠脑组织匀浆,检测脑匀浆液Na+-K+-三磷酸腺苷酶(ATPase),Mg2+-ATPase,Ca2+Mg2+-ATP,Ca2+-ATPase活力;部分脑组织称重后计算脑含水量。结果:脑脉舒康胶囊可以增强Na+-K+-ATPase,Mg2+-ATPase,Ca2+-Mg2+-ATP,Ca2+-ATPase活力,降低MDA含量,提高SOD的活性,减轻脑缺血引起的脑组织间质水肿。结论:脑脉舒康胶囊可提高脑组织ATP能量,稳定离子泵功能,清除氧自由基,缓解脑水肿,具有保护脑细胞的作用。
Objective: To investigate the effect of Naomai Shukang capsule on transient ischemic attack in mice. Methods: Kunming mice were divided into Naomaikang Capsule group (1.8,0.9,0.45 g.kg-1), model group, sham operation and positive drug (Xueshuantong 1.125 g.kg-1, nimodipine 0.03 g · kg-1). The rats in each group were given gavage for 7 days. On the 8th day after administration, the common carotid arteries were ligated for 30 minutes and reperfused for 15 minutes to prepare transient ischemic model ; Blood was collected from the eyes and the activity of superoxide dismutase (SOD) and the content of malondialdehyde (MDA) were measured; some mouse brain tissue homogenates were taken for detection of Na + -K + -ATPase, Mg2 + -ATPase , Ca2 + Mg2 + -ATP, Ca2 + -ATPase activity; some brain tissue after the calculation of brain water content. Results: Naomaikukang capsule can increase the activity of Na + -K + -ATPase, Mg2 + -ATPase, Ca2 + -Mg2 + -ATP and Ca2 + -ATPase, decrease the content of MDA, increase the activity of SOD and relieve the cerebral interstitial edema . Conclusion: Naomai Shukang Capsule can improve brain tissue ATP energy, stabilize ion pump function, scavenge oxygen free radicals, relieve cerebral edema and protect brain cells.
