目的体外观察不同浓度的酒精对肝癌细胞BEL-7402诱导凋亡的作用,为寻求肝癌的治疗提供新思路。方法不同浓度的酒精与肝癌细胞BEL-7402通过体外培养,MTT法检测细胞增殖的变化;Hoechst 33258/PI荧光双染色观察细胞形态学变化;蛋白质免疫印迹(Western blotting)半定量分析Caspase-3和p53和Bax的表达变化。结果药物浓度在100~300 mmol/L的范围显著抑制肝癌细胞增殖,且抑制率随浓度和时间呈依耐性,Caspase-3,Bax和p53的蛋白表达量明显增加。药物浓度在100~300 mmol/L作用48 h,肝癌细胞皱缩,呈现凋亡各期改变,细胞凋亡率显著高于对照组(P<0.05)。结论不同浓度的酒精对肝癌细胞BEL-7402有诱导凋亡的作用,其抑制作用呈浓度和时间依赖性。
Objective To observe the effect of different concentrations of alcohol on inducing apoptosis of hepatoma BEL-7402 cells in vitro and to provide new ideas for the treatment of hepatocellular carcinoma. Methods Different concentrations of alcohol and hepatoma BEL-7402 cells were cultured in vitro and the cell proliferation was detected by MTT assay. Morphological changes were observed by Hoechst 33258 / PI double staining. Western blotting was used to detect the expression of Caspase-3 and p53 and Bax expression changes. Results The drug concentration in the range of 100-300 mmol / L significantly inhibited the proliferation of hepatocellular carcinoma cells. The inhibitory rate was dependent on the concentration and time, and the protein expressions of Caspase-3, Bax and p53 were significantly increased. The drug concentration of 100 ~ 300 mmol / L for 48 h, hepatocellular carcinoma cells shrinkage, showing changes in the apoptotic stage, the apoptotic rate was significantly higher than the control group (P <0.05). Conclusion Different concentrations of alcohol can induce the apoptosis of BEL-7402 hepatoma cells in a concentration-and time-dependent manner.
