目的:研究九节龙皂苷I对小鼠Lewis肺癌和裸鼠肝癌SMMC-7721的抑制作用。方法:C57BL/6小鼠足跖接种稀释3倍的Lewis肺癌细胞悬液20μl制备肺转移模型,灌胃给药14d后,截下荷瘤足,计算原位抑癌率,继续给药至25d,计算肺转移集落数和转移抑制率;BALB/c/nu裸鼠背部皮下接种肝癌SMMC-7721细胞5×106个/ml复制人肝癌实体瘤模型,连续给药16d,动态测量肿瘤体积、体重,画肿瘤生长曲线,实验结束后,剥离瘤块,计算抑瘤率。结果:九节龙皂苷I25mg/kg~100mg/kg对小鼠Lewis肺癌原位癌抑制率和肺转移均分别在40.5%~54.0%和45.4%~69.1%之间,对裸鼠肝癌SMMC-7721实体瘤的抑制率在45.6%~56.3%之间。结论:九节龙皂苷I对Lewis肺癌转移瘤和裸鼠肝癌SMMC-7721均有抑制作用。
OBJECTIVE: To study the inhibitory effect of aconitine I on SMMC-7721 cells in Lewis lung carcinoma and nude mice. Methods: C57BL / 6 mice were inoculated with 20μl suspension of Lewis lung cancer cells which were diluted 3 times to prepare lung metastasis model. After 14 days of intragastric administration, the tumor-bearing foot was cut off and the tumor suppression rate was calculated. The numbers of lung metastasis colony and the metastasis inhibition rate were calculated. The BALB / c / nu nude mice were subcutaneously inoculated with 5 × 106 cells / ml human hepatocellular carcinoma solid tumor model in SMMC-7721 cells on the back for 16 days. The tumor volume, body weight , Draw the tumor growth curve, after the experiment, peel the tumor block, calculate the inhibition rate. Results: The inhibitory rate and lung metastasis of Lewis lung carcinoma in situ were all between 40.5% -54.0% and 45.4% -69.1%, respectively. The inhibitory rates of I25mg / kg ~ 100mg / kg were significantly higher than those of SMMC-7721 The inhibition rates of solid tumors ranged from 45.6% to 56.3%. Conclusion: Gynostemma pentaphylla I inhibits Lewis lung metastasis and SMMC-7721 hepatoma in nude mice.
