目的:探讨卵透明带免疫对卵巢功能的损伤及作用机理。方法:分别以猪卵透明带抗原和缓冲液通过皮内及两后脚掌多点注射免疫Balb/c小鼠,每2w免疫1次,连续免疫3次。经3次免疫后,测定外周血雌二醇浓度和血清抗卵透明带抗体滴度,观察卵巢组织病理形态学及免疫组化,分析IL-1、EGF在卵巢中的表达。结果:免疫小鼠外周血抗ZP抗体滴度明显高于对照组(P<0.01),而雌二醇浓度则明显低于对照组(P<0.01);且二者存在显著负相关(r=-0.85,P=0.001)。卵巢萎缩,闭锁滤泡增加,生长滤泡和黄体数减少,井有炎性细胞浸润;介导滤泡闭锁的IL-1表达增强,而促进滤泡生长的EGF表达受抑。结论:卵透明带免疫使卵巢闭锁滤泡增加,局部免疫凋节紊乱,滤泡耗竭加速,卵巢功能过早衰退。
Objective: To investigate the effect of zona pellucida immunization on ovarian function and its mechanism. Methods: Balb / c mice were immunized with porcine zona pellucida antigen and buffer by multiple injections intradermally and both hind paws. Immunization was performed once every 2 weeks for three times. After 3 immunizations, the peripheral blood estradiol concentration and serum anti-zona pellucida antibody titers were measured. The histopathology and immunohistochemistry of ovarian tissue were observed. The expression of IL-1 and EGF in ovary was analyzed. Results: The titer of anti-ZP antibody in peripheral blood of immunized mice was significantly higher than that of the control group (P <0.01), while the estradiol concentration was significantly lower than that of the control group (P <0.01) (R = -0.85, P = 0.001). Ovarian atrophy, increased atresia follicles, reduced growth follicles and corpus luteum, well inflammatory cell infiltration; IL-1-mediated follicular atresia enhanced expression, and promote follicular growth inhibition of EGF expression. Conclusion: Immunization with zona pellucida increases ovarian atresia follicles, local immune withered disorders, follicular depletion accelerated, premature ovarian failure.
