目的:采用Box-Behnken响应面法优选鳖血柴胡的最佳炮制工艺。方法:以HPLC测得的主要药效成分柴胡皂苷a、d之和为响应指标,采用3因素3水平的Box-Behnken响应面法设计试验,考察鳖血用量、炒制温度、炒制时间对鳖血柴胡炮制工艺的影响。结果:鳖血柴胡的最佳炮制工艺参数为:鳖血用量15%(每100 g柴胡药材拌入15 g鳖血),炒制温度:110℃,炒制时间:15 min。在此最优工艺条件下,柴胡总皂苷含量可达2.109%。结论:Box-Behnken响应面法用于鳖血柴胡炮制工艺考察简单,可靠。
OBJECTIVE: To optimize the processing technology of Bupleurum chinense with Box-Behnken response surface method. Methods: The response factor was determined by the sum of a and d of saikosaponin, the major pharmacologically active ingredient measured by HPLC. The three-factor and three-level Box-Behnken response surface methodology was used to study the effects of the dosage, Effect on the Processing Technology of Bupleurum Chinense. Results: The best processing parameters of turtle Bupleurum were: turtle blood dosage 15% (mixed with 15 g turtle blood per 100 g Bupleurum medicinal materials), frying temperature: 110 ℃, frying time: 15 min. Under the optimum conditions, total saikosaponin content reached 2.109%. Conclusion: The Box-Behnken response surface method is simple and reliable for studying the processing technology of Bupleurum.
