目的:建立同时测定白英中延龄草苷(trillin)和去半乳糖替告皂苷(desgalactotigonin)含量的HPLC-ELSD法。方法:采用Diamonsil-C18色谱柱(4.6 mm×250 mm,5μm),流动相乙腈-10 mmol.L-1醋酸铵溶液52∶48,柱温25℃,流速0.6mL.min-1,进样量20μL,检测器漂移管温度为95℃,氮气流速2.3 L.min-1。结果:延龄草苷和去半乳糖替告皂苷分别在20~200 mg.L-1(r=0.999 8)和10~100 mg.L-1(r=0.999 7)内线性关系良好。延龄草苷的平均回收率为99.4%,RSD为0.90%;去半乳糖替告皂苷的平均回收率为100.3%,RSD 1.1%。结论:该方法准确,重复性良好,适用于白英药材的质量控制。
OBJECTIVE: To establish a HPLC-ELSD method for the simultaneous determination of trillin and desgalactotigonin in white Chinese cabbage. Methods: Diamonsil-C18 column (4.6 mm × 250 mm, 5 μm) was used. The mobile phase was acetonitrile-10 mmol·L-1 ammonium acetate solution 52:48. The column temperature was 25 ℃ and the flow rate was 0.6 mL.min-1. Volume 20 μL, detector drift tube temperature of 95 ° C, nitrogen flow rate 2.3 L.min-1. Results: The results showed that there was a good linear relationship between trillin and galectin for 20-200 mg.L-1 (r = 0.999 8) and 10-100 mg.L-1 (r = 0.999 7) respectively. The average recovery of trillin glycosides was 99.4% with a RSD of 0.90%. The average recoveries of galactosyl trisubstituted saponins were 100.3% and RSD 1.1%. Conclusion: The method is accurate and reproducible, which is suitable for the quality control of Baiying medicinal materials.
