为了探讨陆英作为骨折常用中草药对成骨细胞的影响和相关机理。以陆英提取物氯原酸(chlorogenic acid,简称CHA)对成骨细胞的作用为研究模型,通过提取纯化获得原代成骨细胞,并经细胞活性观察、组织化学染色及荧光定量PCR检测技术,分别对CHA作用于成骨细胞后,检测成骨细胞活性及CHA对成骨细胞生长及Ⅰ型胶原及骨钙蛋白基因表达的影响。结果表明:10μmol/L,100μmol/L CHA能促进成骨细胞钙化结节形成,且能促进I型胶原基因持续增长;在培养第3d时,CHA对骨钙蛋白的表达具有促进作用。陆英有效成分氯原酸能明显增强成骨细胞I型胶原蛋白和骨钙蛋白基因的表达,合成和分泌骨基质,具有一定的成骨活性。
In order to explore Lu Ying as a fracture common Chinese herbal medicine on osteoblasts and related mechanisms. The effect of chlorogenic acid (chlorogenic acid, abbreviation: CHA) on osteoblasts was studied. Primary osteoblasts were obtained by extraction and purification. Cell viability, histochemical staining and fluorescence quantitative PCR The effect of CHA on osteoblasts and the growth of osteoblasts and the expression of type I collagen and osteocalcin genes were examined after osteoblasts were treated with CHA. The results showed that CHA at 10μmol / L and 100μmol / L could promote the formation of calcified nodules and promote the type Ⅰ collagen gene to continue to grow. On the 3rd day of culture, CHA promoted the expression of osteocalcin. Lu Ying active ingredient chlorogenic acid can significantly enhance osteoblast type I collagen and osteocalcin gene expression, synthesis and secretion of bone matrix, with some osteogenic activity.
