目的:探讨NF-κB、MIF与子宫内膜异位症(EMs)发病的关系,及在子宫内膜异位症表达的相关性。方法:采用免疫组化(SABC法)分别检测NF-κB、MIF在35例EMs在位内膜、60例EMs异位内膜及30例正常子宫内膜(对照组)中的表达;ELISA法检测TNF-α、PDTC+TNF-α干预后子宫内膜基质细胞(ESC)上清液中NF-κB、MIF的浓度,并统计分析检测结果。结果:(1)NF-κB、MIF在3组内膜中均有表达,以异位内膜的表达最强;(2)NF-κB、MIF的表达与月经周期无关,其在3组内膜的表达有相关性,相关系数为0.875、0.882、0.927;(3)NF-κB、MIF在经TNF-α、PDTC+TNF-α处理过的ESC上清液中有表达,TNF-α促进二者表达,而PDTC可抑制此作用,且两者表达有相关性,相关系数为0.947。结论:NF-κB、MIF在EMs内膜高表达,MIF可能通过NF-κB途径在EMs发病机制中起作用。
Objective: To investigate the relationship between NF-κB, MIF and the pathogenesis of endometriosis (EMs) and the expression of endometriosis. Methods: The expressions of NF-κB and MIF were detected by immunohistochemistry (SABC method) in 35 eutopic endometrium, 60 ectopic endometrium of EMs and 30 normal endometrium (control group) The concentrations of NF-κB and MIF in the supernatant of the endometrial stromal cells (ESC) after intervention of TNF-α, PDTC + TNF-α were detected, and the results were statistically analyzed. Results: (1) The expressions of NF-κB and MIF in the endometrium of 3 groups were the highest, and the expression of MIF was the strongest in ectopic endometrium. (2) The expression of NF-κB and MIF had no correlation with menstrual cycle, (3) NF-κB and MIF were expressed in supernatant of TNF-α, PDTC + TNF-α-treated ESC supernatant, TNF-αpromoter PDTC could inhibit this effect, and the expression of the two was correlated, the correlation coefficient was 0.947. Conclusion: The expression of NF-κB and MIF is highly expressed in the endometrium of EMs, and MIF may play a role in the pathogenesis of EMs through the NF-κB pathway.
