目的:建立一种同时测定柴胡属植物地上部分6种黄酮类成分含量的HPLC法。方法:采用Hanbon ODS C18色谱柱(150 mm×6 mm,5μm),流动相A为0.1%甲酸水溶液,B为甲醇-乙腈(5∶1),梯度洗脱,检测波长360 nm。结果:建立了芦丁、广寄生苷、水仙苷、槲皮素、山奈素、异鼠李素的HPLC含量测定方法,6种指标性成分在各自浓度范围内线性关系良好(r2>0.999 7);平均回收率分别为98.35%,99.18%,100.95%,99.05%,98.77%和98.25%;方法重复性RSD分别为0.75%,1.02%,1.55%,1.29%,0.45%和0.56%。结论:该法灵敏快捷,适于柴胡属植物中黄酮类成分的分离与测定。
OBJECTIVE: To establish a HPLC method for simultaneous determination of six flavonoid components in the aerial parts of Bupleurum. Methods: Hanbon ODS C18 column (150 mm × 6 mm, 5 μm) was used. The mobile phase A was 0.1% formic acid aqueous solution and the mixture was methanol-acetonitrile (5:1). The detection wavelength was 360 nm. Results: The HPLC method was established for the determination of rutin, glycosides, narcissus, quercetin, kaempferol and isorhamnetin. The linearity of the six index components was good (r2> 0.999 7) The average recoveries were 98.35%, 99.18%, 100.95%, 99.05%, 98.77% and 98.25%, respectively. The repeatability RSDs were 0.75%, 1.02%, 1.55%, 1.29%, 0.45% and 0.56%, respectively. Conclusion: The method is sensitive and fast, suitable for the separation and determination of flavonoids in Bupleurum.
