目的 研究过氧化氢 (H2 O2 )对体外培养的分化期大鼠胚胎神经细胞的氧化损伤以及硒的保护作用。方法 MTT法检测细胞存活率 ,硫代巴比妥酸法测定丙二醛 (MDA)含量 ,黄嘌呤氧化酶法测定超氧化岐化酶(SOD)活性 ,扫描电镜观察细胞膜完整性。结果 H2 O2 抑制分化期神经细胞分裂增殖 ,经过氧化氢作用 2 4h的神经细胞 ,其胞液中SOD活力下降 ,MDA含量升高 ,并表现为浓度 效应关系 ;扫描电镜观察可见细胞膜损伤。细胞培养液中加入一定浓度硒可有效地减轻H2 O2 引起的神经细胞损伤。结论 硒对H2 O2 引起的神经细胞氧化损伤可能有明显的保护作用。
Objective To study the oxidative damage of hydrogen peroxide (H2O2) to cultured rat embryonic neural cells in vitro and the protective effect of selenium. Methods Cell viability was measured by MTT assay. Malondialdehyde (MDA) content was measured by thiobarbituric acid method. Superoxide dismutase (SOD) activity was measured by xanthine oxidase method and cell membrane integrity was observed by scanning electron microscopy. Results H 2 O 2 inhibited the proliferation and differentiation of neural cells during differentiation. After 24 h of H 2 O 2 administration, H2O2 decreased SOD activity and MDA content in the cytoplasm, and showed a concentration-dependent effect. Cell membrane damage was observed by scanning electron microscopy. Adding selenium to cell culture solution can effectively reduce the damage of neurons caused by H2O2. Conclusion Selenium may have a significant protective effect on H2O2-induced neuronal oxidative damage.
