目的:探讨补肾活血中药对双侧卵巢摘除引起的骨质疏松大鼠mGluR5的影响。方法:取3月龄SPF级雌性SD大鼠70只,随机分为造模组(60只)行双侧卵巢摘除和假手术组(Sham group,10只)行假手术,术后30 d将造模组动物随机分为6组(10只/组):补肾活血方组(BSHX group)、龟鹿二仙胶组(GLEX group)、二仙汤组(EX group)、仙灵骨葆组药物对照组(XLGB group)及雌二醇药物对照组(EV group)各自按1 mL/100 g体质量给于相应的药物灌胃,假手术组(Sham group)和模型组(Model group)各自灌服等量的生理盐水。连续给药12周后测定各组大鼠血清中Ⅰ型胶原羧基端肽(β-Crosslaps)、总Ⅰ型胶原氨基端延长肽(P1NP)、TRAP(抗酒石酸酸性磷酸酶)的含量;测定大鼠的骨密度;用Western-blot方法检测股骨骨骺Glu、mGluR5蛋白表达。结果:与Sham组比较,Model组大鼠骨密度、血液PINP浓度、Glu表达均显著降低(P<0.01),β-Crosslaps、TRAP浓度及mGluR5表达显著增高(P<0.01)。与Model组比较,各治疗组大鼠骨密度、PINP、Glu蛋白表达显著增高(P<0.05),β-Crosslaps、TRAP浓度及mGluR5表达显著降低(P<0.01),补肾活血组骨密度及Glu表达高于其他各治疗组。结论:补肾活血能有效防止去卵巢模型大鼠骨质疏松的形成,可能通过调节骨内谷氨酸进一步介导骨重建而发挥作用。
Objective: To investigate the effect of Bushen Huoxue Chinese medicine on mGluR5 in rats with osteoporosis induced by ovariectomy. Results: Compared with Sham group, BMD, blood PINP concentration and Glu expression in model group were significantly decreased (P <0.01), and the levels of β-Crosslaps, TRAP and mGluR5 were significantly increased (P <0.01). Compared with model group, the BMD, PINP and Glu protein expression in each treatment group were significantly increased (P <0.05), the levels of β-Crosslaps, TRAP and mGluR5 were significantly decreased (P <0.01) Expression higher than the other treatment groups. Conclusion: Bushenhuoxue can effectively prevent the formation of osteoporosis in ovariectomized rats and may play a role in the further regulation of bone remodeling by regulating intraosseous glutamate.
