[目的]探讨金针菇发酵液对血瘀模型大鼠血管内皮细胞及血小板功能的影响.[方法]采用肾上腺素结合冰浴的方法制作大鼠血瘀模型,灌胃给予金针菇发酵液后观察血管组织形态学改变.采用流式细胞仪检测血小板膜糖蛋白CD62p水平,利用ELISA法测定血浆p-选择素、6-酮-前列腺素E_(1_α)(6-keto-PGE_(1_α))、血栓素B_2(TXB_2)和血管紧张素Ⅱ(AngⅡ)水平,采用硝酸还原酶法检测血浆一氧化氮含量.[结果]与模型对照组比较,金针菇发酵液低、高剂量组血管组织结构规整,内膜连续,未见炎性细胞浸润;金针菇发酵液高剂量组6-keto-PGE_(1_α)水平明显升高,而血小板膜糖蛋白CD62p、血浆p-选择素、TXB_2和AngⅡ水平均明显下降(P<0.05,P<0.01).[结论]金针菇发酵液具有保护血管内皮细胞及抗血小板活化的作用.
[Objective] To explore the effect of Flammulina velutipes fermentation broth on vascular endothelial cells and platelet function in blood stasis model rats. [Methods] The blood stasis model was made by adrenaline combined with ice bath. Morphological changes were detected by flow cytometry platelet membrane glycoprotein CD62p levels by ELISA assay of plasma p-selectin, 6-keto-prostaglandin E_ (1_α) (6_ keto-PGE_ (1_α)), thromboxane B_2 (TXB_2) and angiotensin Ⅱ (AngⅡ) were measured by enzyme-linked immunosorbent assay (ELISA). [Results] Compared with the model control group, The levels of 6-keto-PGE_ (1_α) in the high-dose group of Flammulina velutipes fermentation broth were obviously increased, while the levels of platelet membrane glycoprotein CD62p, plasma p-selectin, TXB_2 and AngⅡ were significantly decreased <0.05, P <0.01). [Conclusion] Flammulina velutipes fermentation broth has the function of protecting vascular endothelial cells and anti-platelet activation.
