目的建立超高效液相色谱(UPLC)法同时测定肿节风超微粉中异嗪皮啶和迷迭香酸的含量,并比较不同粒度肿节风有效成分溶出率差异。方法采用UPLC法,Eclipse Plus C18(2.1 mm伊50 mm,1.8滋m),流动相为甲醇(A)-0.1%磷酸(B),梯度洗脱,流速为0.4 mL·min-1,柱温为35益,紫外检测波长为342 nm。结果异嗪皮啶和迷迭香酸分别在9.97耀59.84滋g·mL-1(r=0.99995),10.90耀60.14滋g·mL-1(r=0.99935)范围内线性关系良好,平均回收率(n=6)分别为95.16%(RSD=2.15%),100.31%(RSD=2.29%)。结论所建立的UPLC法操作简便快捷、经济、准确可靠、分离度好,可为肿节风药材有效成分分析和质量控制提供新的方法。
OBJECTIVE To establish a method for the simultaneous determination of isofraxidin and rosmarinic acid in superfine powder of swollenang section by ultra performance liquid chromatography (UPLC) and compare the dissolution rates of effective components in different size swollen sections. Methods UPLC method was used. The mobile phase consisted of Eclipse Plus C18 (2.1 mm × 50 mm, 1.8 μm) and mobile phase consisted of methanol (A) -0.1% phosphoric acid (B) with gradient elution at a flow rate of 0.4 mL · min- 35 benefits, UV detection wavelength of 342 nm. Results The isoindoline and rosmarinic acid showed good linearity in the ranges of 9.97 ± 59.84 μg · mL-1 (r = 0.99995) and 10.90 ± 60.14 μg · mL-1 (r = 0.99935) respectively. The average recovery (n = 6) were 95.16% (RSD = 2.15%) and 100.31% (RSD = 2.29%) respectively. Conclusion The established UPLC method is simple, rapid, economical, accurate and reliable, and has good resolution. It can provide a new method for the active ingredient analysis and quality control of the herbal medicine of Arthritis.
