应用放射配体结合法检测不同病期被动型Heymann肾炎(PHN)病鼠肾、肝胞液糖皮质激素受体(GR),同时测定相应病期血浆皮质酮(B)浓度。结果:病鼠第2周末肾胞液GR最大结合容量(R_0)较正常大鼠减少(P<0.05);3周末进一步减少(P<0.005);4周末R_0已回升,与正常大鼠差异不显著(P>0.05)。这种动态变化与疾病的发生、发展及转归相一致。肾胞液GR的R_0与蛋白量呈负相关(P<0.005),与血肌酐值亦呈负相关(P<0.01)。而肝胞液GR的R_0及血浆B的浓度各病期与正常对照组均无显著差异(P>0.05)。结果提示:PHN大鼠肾组织局部GR减少与PHN的发病存在密切联系。
Radioligand binding assay was used to detect glucocorticoid receptor (GR) in renal and hepatic cytokines in passive Heymann nephritis (PHN) and plasma corticosterone (B) concentrations in corresponding phase. Results: Compared with normal rats, the maximum binding capacity (R_0) of renal GR in BALF at the second weekend in rats decreased (P <0.05), decreased further at the third week (P <0.005) There was no significant difference in rats (P> 0.05). This dynamic change is consistent with the occurrence, development and outcome of the disease. Renal cytosolic GR R_0 negative correlation with protein levels (P <0.005), and serum creatinine values were also negatively correlated (P <0.01). However, there was no significant difference in the levels of R_0 and plasma B in liver cells between normal control group and control group (P> 0.05). The results suggest that there is a close relationship between the decrease of local GR in PHN rats and the incidence of PHN.
