目的探讨魔芋甘露低聚糖(konjac mannan oligosaccharides,KMOS)对高脂血症大鼠的治疗作用。方法 56只雄性SD大鼠随机分为5组,阴性对照组、模型组、KMOS低剂量实验组(KMOS-L组)、KMOS中剂量实验组(KMOS-M组)、KMOS高剂量实验组(KMOS-H组)。除阴性对照组外的四组大鼠均给予高脂饲料喂养,第5周末,采集大鼠尾血检测血脂水平筛选出高脂血症大鼠。3组实验组高脂血症大鼠分别给予低、中、高剂量40 mg/100 g、80 mg/100 g、160 mg/100 g KMOS每日灌胃,模型组大鼠给予生理盐水每日灌胃,每周称体质量,灌胃期间均继续给予高脂饲料喂养,第10周末断头处死大鼠,取全血,离心后待测。检测大鼠血清总胆固醇(total cholesterol,TC)、甘油三酯(triglyceride,TG)、高密度脂蛋白胆固醇(high density lipoprotein cholesterol,HDL-C)、低密度脂蛋白胆固醇(low density lipoprotein cholesterol,LDL-C)含量,空腹血糖(glucose,Glu)、胰岛素(insulin,INS)含量,血清丙二醛(malondialdehyde,MDA)、还原型谷胱甘肽(glutathione,GSH)、超氧化物歧化酶(superoxide dismutase,SOD)含量。结果第5周到第10周,4组高脂饲料组大鼠体质量均显著高于对照组(P<0.05);5周末,4组高脂饲料组大鼠的血清TC,Glu,INS和MDA水平显著高于对照组(P<0.05),HDL-C水平显著低于对照组(P<0.05);10周末,KMOS-M和KMOS-H组血清TC和MDA含量较模型组显著降低(P<0.05),三组KMOS组血清TG含量较模型组显著降低(P<0.05),KMOS-H组空腹血糖较模型组显著降低(P<0.05)。结论 KMOS能降低高脂血症大鼠血清TC,TG,Glu和MDA含量,对其有一定治疗作用。
Objective To investigate the therapeutic effect of konjac mannan oligosaccharides (KMOS) on hyperlipidemic rats. Methods 56 male Sprague-Dawley rats were randomly divided into 5 groups: negative control group, model group, KMOS low dose group (KMOS-L group), KMOS middle dose group (KMOS-M group), KMOS high dose group KMOS-H group). Except for the negative control group, four groups of rats were fed with high-fat diet. At the end of the fifth week, rats were collected to detect the level of lipids in the tail blood and the hyperlipidemic rats were selected. Rats in the three experimental groups were given low, medium and high doses of KMOS 40 mg / 100 g, 80 mg / 100 g and 160 mg / 100 g KMOS orally daily. Rats in the model group were given normal saline daily Gavage, weekly body mass, gavage were continued to give high-fat diet, the first 10 weeks of decapitation rats were sacrificed, take whole blood, centrifuged to be tested. Serum levels of total cholesterol (TC), triglyceride (TG), high density lipoprotein cholesterol (HDL-C), low density lipoprotein cholesterol (LDL) C content, fasting glucose (Glu), insulin (INS), serum malondialdehyde (MDA), glutathione (GSH), superoxide dismutase dismutase, SOD) content. Results From the 5th week to the 10th week, the body weight of four groups of high-fat diet rats were significantly higher than that of the control group (P <0.05). At the end of the 5th week, the serum TC, Glu, INS and MDA (P <0.05). The levels of serum TC and MDA in KMOS-M and KMOS-H groups were significantly lower than those in the model group at the end of the 10 th week (P <0.05) <0.05). The levels of serum TG in the three groups of KMOS rats were significantly lower than those in the model group (P <0.05). The fasting blood glucose of KMOS-H rats was significantly lower than that in the model group (P <0.05). Conclusion KMOS can reduce the content of TC, TG, Glu and MDA in serum of hyperlipidemic rats, which may have a certain therapeutic effect on it.
