3种不同类型的细毡毛忍冬遗传关系的ISSR分析

目的探索3种不同类型细毡毛忍冬在分子水平上的差异。方法利用ISSR分子标记分析3种不同类型细毡毛忍冬的48份材料的遗传多样性。结果从100条ISSR引物中筛出8条多态性高、条带清晰的引物,对DNA样品进行了PCR扩增,得到68个可统计的条带,其中60条具有多态性的条带,占88.2%。POPGENE 1.32软件统计结果显示:3种不同类型细毡毛忍冬居群间的遗传一致度较高,范围为0.9549~0.9611;遗传距离较小,范围为0.0397~0.0462。3个居群总的遗传多样性(Ht)为0.2939,居群内的遗传多样性(Hs)为0.2735,表明总的遗传变异主要来自于居群内。根据总的遗传多样性和居群内遗传多样性计算不同居群间的分化水平(Gst),所分析的3个细毡毛忍冬居群间的Gst为0.0694,说明居群间的分化水平很低。基因流Nm为6.7036,表明居群间有很强的基因交流。结论影响3种细毡毛忍冬形态差异的因素可能并非遗传因素,而是环境、人文等其他因素。 Objective To explore the molecular differences of three different types of Fritillaria nivalis. Methods The ISSR molecular markers were used to analyze the genetic diversity of 48 accessions from 3 different types of Lonicera macranthoides. Results Eighty hundred ISSR primers were screened out of eight highly polymorphic and well-defined primers, and the DNA samples were PCR-amplified to obtain 68 statistically significant bands, of which 60 bands were polymorphic , Accounting for 88.2%. The results of POPGENE 1.32 software showed that the genetic identity among populations of the three species was higher than that of other populations, ranging from 0.9549 to 0.9611. The genetic distance was relatively small, ranging from 0.0397 to 0.0462.3. (Ht) was 0.2939, and the genetic diversity (Hs) within the population was 0.2735, indicating that the total genetic variation mainly came from within the population. According to the total genetic diversity and genetic diversity among populations, the level of differentiation (Gst) among different populations was calculated. The Gst among three populations analyzed was 0.0694, indicating that the level of differentiation among populations was very high low. Gene flow Nm 6.7036, indicating that there is a strong genetic exchange among populations. Conclusion The factors affecting the morphological differences of the three species of Fritillaria nivalis may not be genetic factors, but other factors such as environment, humanities and so on.
