目的:探讨合并HCV感染对HIV-1感染者特异性杀伤性CD4n + T细胞免疫应答的影响。n 方法:选取2015年9月至2016年5月在广州市第八人民医院就诊的49例HIV-1感染者,根据是否合并HCV感染,分为HIV-1单纯感染组(26例)和HIV-1合并HCV感染组(23例)。以HIV-1 P24蛋白上的氨基酸序列人工合成的12个重叠肽段组成肽段库,体外刺激已接受HAART的外周CD4n + T细胞,采用酶联免疫斑点试验方法检测特异性CD4n + T细胞分泌IFN-γ分泌细胞个数,根据斑点形成细胞(SFC)数计算抗原特异性CD4n + T细胞应答频率。以1×10n 6个细胞产生的SFC>50,且大于各自非特异性刺激对照孔SFC的3倍作为HIV-1抗原特异性CD4n + T细胞阳性反应。n 结果:HIV-1单纯感染组的特异性杀伤性CD4n + T细胞阳性应答率为80.8%,应答频率为250(100,700)SFC/10n 6个细胞,均高于HIV-1合并HCV感染组的52.2%和50(0,250)SFC/10n 6个细胞,组间比较差异具有统计学意义(n χn 2=4.538, n Z=-2.432, n P均<0.05)。n 结论:HIV-1感染者合并HCV感染可能抑制特异性杀伤性CD4n + T细胞反应。n “,”Objective:To investigate the effect of HIV-1 co-infection with HCV on immune responses of HIV-1 specific cytotoxic CD4n + T cells.n Methods:There were 49 confirmed HIV-1 infected patients selected at Guangzhou Eighth People’s Hospital from September 2015 to May 2016. According to whether they were co-infected with HCV, 26 and 23 patients were assigned to HIV-1 infection group and HIV-1/HCV co-infection group, respectively. The peripheral CD4n + T cells of the patients who had received HAART were stimulated n in vitro by a peptide pool containing 12 overlapping peptides that synthesized from the amino acid sequence of HIV-1 P24 protein. The number of IFN-γ-secreting cells secreted by specific CD4n + T cells was detected by enzyme-linked immunospot assay. The response frequency of antigen-specific CD4n + T cells was calculated by the number of spot-forming cells (SFC) . If SFC produced by 1×10n 6 cells was more than 50, and 3 times greater than that in respective non-specific stimulus control wells, it was defined as positive response of HIV-1 antigen-specific CD4n + T cell.n Results:The positive response rate of specific cytotoxic CD4n + T cells in HIV-1 infection group was 80.8%, and the response frequency was 250 (100, 700) SFC/10n 6 cells, which were both higher than those of 52.2% and 50 (0, 250) SFC/10n 6 cells in HIV-1/HCV co-infection group. The differences between the two groups were statistically significant (n χ2=4.538, n Z=-2.432, n P both <0.05) .n Conclusions:HCV co-infection in HIV-1 infected individuals may inhibit responses of specific cytotoxic CD4n + T cells.n
