[目的]研究组织蛋白酶B(CathepsinB)在紫杉醇诱导胃癌SGC-7901细胞凋亡中的作用。[方法]用0.1~0.5μmol/L紫杉醇作用于SGC-7901细胞,另一组再加入CathepsinB特异性抑制剂CA-074(Me)对照;再用0.3μmol/L紫杉醇对SGC-7901细胞分别作用0h、6h、12h、24h、36h、48h,另一组再加入CA-074(Me),通过形态学观察及流式细胞术检测细胞凋亡率的变化。[结果]①随着紫杉醇浓度的增高和作用时间的延长,SGC-7901细胞的凋亡率明显增高;②加入CA-074(Me)后,细胞凋亡率明显下降。[结论]CathepsinB在紫杉醇诱导的细胞凋亡中有着相对重要的作用。
[Objective] To investigate the role of Cathepsin B in taxol-induced apoptosis of gastric cancer SGC-7901 cells. [Method] SGC-7901 cells were treated with 0.1 ~ 0.5μmol / L paclitaxel and CA-074 (Me) control was added as Cathepsin B specific inhibitor in another group. SGC-7901 cells were treated with 0.3μmol / L paclitaxel 0h, 6h, 12h, 24h, 36h, 48h. The other group was added with CA-074 (Me), and the change of apoptosis rate was detected by morphological observation and flow cytometry. [Results] ① The apoptosis rate of SGC-7901 cells was significantly increased with the increase of paclitaxel concentration and the prolongation of action time. ② The apoptosis rate of CA-074 (Me) cells was significantly decreased after addition of paclitaxel. [Conclusion] Cathepsin B plays a relatively important role in paclitaxel-induced apoptosis.
