目的 探讨银杏内酯B(GB)对牛主动脉平滑肌细胞 (SMC)增殖的影响及可能机制。方法 细胞的增殖由3H 胸腺嘧啶核苷掺入试验确定 ,应用流式细胞术测定细胞周期 ,用MTT法和乳酸脱氢酶活性检测观察细胞代谢的变化。结果 无论是否用血管紧张素II诱导 ,GB与银杏内酯A和B的混合物均呈浓度依赖性地抑制SMC的增殖 ,其机制与阻止细胞进入有丝分裂期有关。结论 GB可抑制动脉壁SMC的增殖 ,该作用不完全取决于血小板活化因子受体的拮抗活性
Objective To investigate the effects of ginkgolide B (GB) on the proliferation of bovine aortic smooth muscle cells (SMCs) and its possible mechanism. Methods Cell proliferation was determined by 3H thymidine incorporation assay. Cell cycle was determined by flow cytometry. MTT assay and lactate dehydrogenase activity assay were used to observe the changes of cell metabolism. Results Both GB and Ginkgolide A and B inhibited proliferation of SMCs in a concentration-dependent manner, regardless of whether they were induced with angiotensin II, by a mechanism that is associated with preventing cells from entering the mitotic phase. Conclusion GB can inhibit the proliferation of arterial wall SMC, the effect does not depend entirely on the activity of platelet-activating factor receptor antagonist
