目的 :探讨天然产物虎眼万年青多糖 (S3)对 T细胞及其细胞因子 IL - 2 m RNA表达的影响 ,进一步从细胞和分子水平探讨其免疫调节作用机制 ,为开发这一天然产物提供理论依据。方法 :利用流式细胞术 ,检测S3对 T淋巴细胞亚群 CD3、 CD4、 CD8、 CD4 / CD8阳性细胞百分率的影响 ,并采用 RT- PCR方法检测 IL - 2 m RNA的表达水平。结果 :各剂量组 CD3、CD4阳性细胞百分率均有上升趋势 ,极高剂量组 CD3、CD4与对照组比较差异有显著性 (P<0 .0 0 1) ,CD8有下降趋势 ,各剂量组均能显著提高 CD4 / CD8阳性细胞百分率 (P<0 .0 0 1) ;极高剂量组和高剂量组可明显促进脾细胞中细胞因子 IL- 2的 m RNA表达 ,使表达量增加 (P<0 .0 1,P<0 .0 5 )。结论 :S3具有较强的增强机体多种免疫功能的作用。
Objective: To investigate the effect of the natural product of tiger eye evergreen polysaccharide (S3) on the expression of T-cells and its cytokine IL-2 mRNA, and to further explore its immunoregulatory mechanism from the cellular and molecular levels, providing a theoretical basis for the development of this natural product. . Methods : Flow cytometry was used to detect the effect of S3 on the percentage of CD3, CD4, CD8 and CD4/CD8 positive cells in T lymphocyte subsets. The expression of IL-2 mRNA was detected by RT-PCR. RESULTS: The percentage of CD3 and CD4 positive cells in each dose group had an upward trend. The CD3 and CD4 in the very high dose group were significantly different from those in the control group (P<0.01). CD8 had a decreasing trend. Each dose group was It can significantly increase the percentage of CD4/CD8 positive cells (P<0.01). Very high dose group and high dose group can significantly promote the expression of cytokine IL-2 in the spleen cells and increase the expression level (P< 0 .0 1,P<0 .0 5 ). Conclusion :S3 has a strong role in enhancing various immune functions of the body.
