目的 :研究急淋白血病患儿红细胞内 6 -MP 3种代谢产物的含量情况。方法 :应用反相高效液相色谱技术 ,采用外标法 ,样品经乙酸苯汞加合物提取 ,色谱柱为DupondC18(10 μm ,4 6mm× 2 5 0mm) ,柱温 15℃~ 2 0℃ ,流动相为甲醇 -水 (5∶95 ) ,流速 0 9mL·min-1,检测波长 330和 30 0nm。结果 :红细胞内 6 -TGN、 6 -TIMP、 6 -MeMP的浓度在 5~ 2 0 0 0ng/ 8× 10 8红细胞范围内线性关系良好 ,rs=0 885 5~ 0 9871,天内RSD为0 2 %~ 1 2 % ,天间RSD为 1 1%~ 2 1%。结论 :本方法简便、快速、准确 ,适用于 6 -MP细胞内药理学研究
Objective: To study the content of 6-MP 3 metabolites in erythrocytes of children with acute leukemia. Methods: Reversed-phase high performance liquid chromatography (RP-HPLC) was used. The sample was extracted with phenylmercuric acetate by external standard method. The column was Dupond C18 (10 μm, 46 mm × 250 mm) with column temperature 15 ℃ ~ 20 ℃ , The mobile phase was methanol-water (5:95), the flow rate was 9 mL · min-1 and the detection wavelength was 330 and 300 nm. Results: The concentration of 6-TGN, 6-TMPMP and 6-MeMP in erythrocytes was good in the range of 5-20 000 ng / 8 × 10 8 erythrocytes, rs = 0 885 5-0 9871, and RSD was 0 2 % ~ 12%, inter-day RSD 11% ~ 21%. Conclusion: The method is simple, rapid and accurate and is suitable for 6-MP intracellular pharmacology research
