目的研究长期不同强度运动对大鼠胫骨前肌HSL、Perilipin基因表达及血清FFA含量的影响。方法 46只雄性SD大鼠,适应练习时淘汰6只。将40只大鼠随即分为安静对照组(C组)、中等耐力运动组(M组)、大强度运动组(N组)、抗阻训练组(R组)。10周训练结束后采用生化方法测定各组大鼠血清FFA含量,RT-PCR法测定ATGL、HSL基因表达。结果与C组相比,运动各组血清中FFA含量增加(P﹤0.05,P﹤0.01);R组、N组HSL基因表达上调(P﹤0.05),其中R组基因表达量最多;R组、N组Perilipin基因表达下调(P﹤0.05),其中以R组基因表达最少。结论抗阻、耐力训练均可以导致骨骼肌HSL基因表达上调,Perilipin基因表达下调,血清FFA含量增加。抗阻、大强度耐力训练显著提高HSL基因表达水平,降低Perilipin基因表达水平,提示Perilipin基因表达的下调似乎可以促进HSL基因表达上调,提高骨骼肌脂肪水解能力。
Objective To study the effect of long-term exercise with different intensity on the expression of HSL and Perilipin in rat tibialis anterior muscle and serum FFA content. Methods Forty-six male Sprague-Dawley rats were used to get rid of 6 rats. Forty rats were divided into quiet control group (C group), middle endurance exercise group (M group), intensive exercise group (N group) and resistance training group (R group). After 10 weeks of training, serum FFA levels were determined by biochemical methods. ATGL and HSL gene expression were determined by RT-PCR. Results Compared with group C, the content of FFA in each group increased (P <0.05, P <0.01). The expression of HSL gene in group R and group N was up-regulated (P <0.05) , Perilipin gene expression was down-regulated in N group (P <0.05), and the expression of Perilipin was the lowest in group R. Conclusion Both resistance and endurance training can lead to up-regulation of skeletal muscle HSL gene expression, down-regulation of Perilipin gene expression and increase of serum FFA content. Resistance and intensive endurance training significantly increased HSL gene expression and decreased Perilipin gene expression, suggesting that the down-regulation of Perilipin gene expression may seem to promote HSL gene expression and enhance the capacity of skeletal muscle lipolysis.
