目的研究DNA结合分化抑制蛋白1(Id-1)在结直肠腺癌组织中的表达并探讨其与致癌的相关性。方法选取我院结直肠癌和正常黏膜组织各50例,用逆转录聚合酶链反应法和免疫印迹法检测2组中Id-1 mRNA和蛋白质的表达情况,并用免疫组化法检测Id-1在2组中的表达。结果结直肠癌组织中,Id-1mRNA的表达水平(0.96±0.03)较正常组织(0.20±0.04)明显增高(P<0.01);Id-1蛋白在结直肠癌组织中的表达水平(0.82±0.04)亦较正常组织(0.31±0.02)明显增高(P<0.01);Pearson相关分析显示,Id-1mRNA和蛋白水平呈明显正相关(P<0.01)。Id-1在结直肠癌组织组阳性表达率是72.00%明显高于正常黏膜的24.00%(P<0.01)。结论 Id-1的特异性高表达与结直肠腺癌的发生与发展具有较高的相关性。
Objective To investigate the expression of DNA-binding inhibitor of differentiation 1 (Id-1) in colorectal adenocarcinoma and to explore its relationship with carcinogenesis. Methods 50 cases of colorectal cancer and normal mucosa were selected in our hospital. The expression of Id-1 mRNA and protein in the two groups were detected by RT-PCR and Western blotting. The expression of Id-1 Expression in two groups. Results The expression level of Id-1 mRNA in colorectal cancer tissues (0.96 ± 0.03) was significantly higher than that in normal tissues (0.20 ± 0.04) (P <0.01) 0.04) was also significantly higher than that of normal tissue (0.31 ± 0.02) (P <0.01). Pearson correlation analysis showed that there was a significant positive correlation between Id-1mRNA and protein level (P <0.01). The positive expression rate of Id-1 in colorectal cancer tissue was 72.00%, which was significantly higher than that in normal mucosa (P <0.01). Conclusion The high expression of Id-1 is associated with the occurrence and development of colorectal adenocarcinoma.
