目的研究db/db自发性II型糖尿病(T2DM)小鼠的饲育方法、胰岛的分离纯化及其胰岛功能的检测。方法杂合子(+/db)采用自然交配,饲养于屏障环境中,常规全价营养饲料喂养。采用胰总管逆行灌注的方法分离胰腺,Ficoll-400密度梯度离心法纯化胰岛。结果 db/db小鼠占繁殖总数的1/2,其中以第4胎产子数最多,4胎后逐渐下降。纯化后的胰岛在倒置显微镜下呈圆形或卵圆形,纯度>90%,体外释放试验提示胰岛B细胞功能较差。结论屏障环境中db/db小鼠稳定繁殖,可成功分离出小鼠的胰岛并将其纯化。
Objective To study the breeding method, isolation and purification of pancreatic islets and the detection of islet function in db / db spontaneous type 2 diabetes mellitus (T2DM) mice. Methods Heterozygotes (+ / db) were naturally bred and housed in a barrier environment, fed with conventional full-price nutritive feeds. Pancreas were isolated by retrograde perfusion of pancreatic duct and islets were purified by Ficoll-400 density gradient centrifugation. Results db / db mice accounted for 1/2 of the total number of reproductive, of which the highest number of births in the fourth child, 4-fetal decline. The purified islets were round or oval under an inverted microscope with a purity of> 90%. In vitro release test indicated that islet B cells were poorly functional. Conclusion db / db mice are stable in the barrier environment and the islets of mice can be successfully isolated and purified.
