目的 应用荧光定量RT-PCR(Fluorogenic quantitative reverse transcriptase polymerasechain reaction,RT-PCR)检测肺癌外周血中微转移情况。方法 用荧光定量RT-PCR方法检测57例肺癌患者外周血CK19mRNA表达。结果 57例肺癌患者外周血中CK19mRNA检测阳性19例,阳性率33.3%,统计结果显示外周血CK19mRNA的表达与性别、年龄、病理类别无关,在不同分化程度、淋巴结转移状态、临床分期间有显著差异。结论 以CK19mRNA为分子标记,用荧光定量RT-PCR法检测肺癌患者外周血微转移具有很高的灵敏度,有助于亚临床转移的早期发现。
Objective To detect the micrometastasis in peripheral blood of patients with lung cancer by Fluorogenic quantitative reverse transcriptase polymerase chain reaction (RT-PCR). Methods The expression of CK19mRNA in peripheral blood of 57 patients with lung cancer was detected by fluorescence quantitative RT-PCR. Results The positive rate of CK19 mRNA in peripheral blood of 57 patients with lung cancer was 19 (positive rate was 33.3%). The statistical results showed that the expression of CK19mRNA in peripheral blood had no relation with gender, age and pathological type, and there was significant difference in different degree of differentiation, lymph node metastasis and clinical stage difference. Conclusion Using CK19mRNA as a molecular marker, the detection of micrometastases in peripheral blood of patients with lung cancer by fluorescence quantitative RT-PCR is highly sensitive and contributes to the early detection of subclinical metastasis.
