目的:研究c-myc蛋白和MMP-9在受压静脉管壁的表达,初步探讨静脉受压后血管重塑的分子机制。方法:42只SD大鼠随机分为对照组(21只)和实验组(每时间点各3只,共21只),用5注射针头辅助作部分髂静脉结扎造模。分别在0.5、12、24、48、72h、7和14d取材,明胶液包埋后冰冻切片,用免疫组化SP法检测c-myc蛋白和MMP-9的表达,采用计算机图像分析系统测定切片的平均积分光密度(IOD)。结果:对照组和正常静脉管壁均未见c-myc蛋白、MMP-9表达,实验组c-myc蛋白从12h开始表达,MMP-9从24h开始表达上调,并逐渐增强,IOD逐渐升高,与对照组比较,差异具有显著性(P<0.05)。7d时两者表达同时达到高峰。c-myc蛋白与MMP-9在受压静脉管壁的表达呈正相关(r=0.7532,P<0.05)结论:c-myc基因产物和MMP-9在受压静脉管壁表达增强,两者在受压静脉管壁重塑过程起着重要作用;c-myc基因和MMP-9的表达具有相关性。
OBJECTIVE: To study the expression of c-myc and MMP-9 on the surface of compressed venous vessels and to explore the molecular mechanism of vascular remodeling after venous compression. Methods: Forty-two SD rats were randomly divided into control group (n = 21) and experimental group (n = 3 each). Fifty rats were used to establish part of iliac vein ligation. The samples were harvested at 0.5, 12, 24, 48, 72h, 7 and 14d respectively. After the gelatin solution was embedded, the frozen sections were harvested. The expressions of c-myc and MMP-9 were detected by immunohistochemical SP method. Average integrated optical density (IOD). Results: The expression of c-myc protein and MMP-9 were not found in the control group and the normal vein. The expression of c-myc protein in the experimental group began at 12 h, the expression of MMP-9 was up-regulated from 24h, and gradually increased , Compared with the control group, the difference was significant (P <0.05). At 7d, both of them reached the peak at the same time. The expression of c-myc protein was positively correlated with the expression of MMP-9 in the compressed venous wall (r = 0.7532, P <0.05) .Conclusion: The expressions of c-myc and MMP- Pressure vessel wall remodeling process plays an important role; c-myc gene and MMP-9 expression is correlated.
