目的研究大肠癌患者K-ras基因的突变情况及其与临床病理特征间的关系。方法应用RT-PCR方法检测148例大肠癌患者组织标本K-ras基因第12和13密码子的突变情况,并与其临床病理特征进行对照。结果 148例大肠癌患者K-ras基因突变率为31.8%(47/148)。K-ras基因第12和13密码子突变在不同性别、肿瘤直径、大体形态、肿瘤部位、远处及淋巴结转移方面差异无统计学意义(P>0.05),在不同年龄与不同临床分期上差异有统计学意义(P<0.05)。结论 RT-PCR可准确检测K-ras基因突变情况,K-ras基因突变与患者年龄、肿瘤临床分期关系密切。
Objective To investigate the mutation of K-ras gene in patients with colorectal cancer and its relationship with clinicopathological features. Methods Mutations of codons 12 and 13 of K-ras gene in 148 specimens of colorectal cancer tissues were detected by RT-PCR and compared with the clinicopathological features. Results The mutation rate of K-ras gene in 148 cases of colorectal cancer was 31.8% (47/148). The mutation of codons 12 and 13 of K-ras gene had no significant difference in different gender, tumor diameter, general morphology, tumor location, distant location and lymph node metastasis (P> 0.05), but different in different age and clinical stages There was statistical significance (P <0.05). Conclusion RT-PCR can accurately detect the K-ras gene mutation. The mutation of K-ras gene is closely related to patient’s age and clinical stage.
