目的:探讨急性高糖对缺血再灌注后大鼠肾组织髓过氧化物酶(MPO)和环氧合酶2(Cox-2)表达的影响。方法:成年雄性SD大鼠随机分为正常血糖假手术(NG-Sham)组、正常血糖缺血再灌注(NG-I/R)组、高糖假手术(HG-Sham)组、高糖缺血再灌注(HG-I/R)组4组(n=8)。大鼠切除右侧肾脏,夹闭对侧肾蒂45 min再灌注24 h制备肾缺血再灌注模型。大鼠急性高糖模型为术前45 min通过腹腔注射25%葡萄糖3 g·kg-1制备。比色法检测大鼠肾脏MPO,RT-PCR法检测肾脏Cox-2-mRNA,Western blotting检测肾脏Cox-2蛋白表达。结果:与Sham组相比,I/R组MPO活性,Cox-2-mRNA、Cox-2蛋白表达均显著增高(P<0.05),而HG-I/R组的MPO活性,Cox-2-mRNA、Cox-2蛋白表达又明显高于NG-I/R组(P<0.05)。结论:急性高糖可以增加缺血再灌注后大鼠肾组织MPO活性,增加Cox-2的表达水平,加重肾缺血及再灌注所引发的炎症损伤。
Objective: To investigate the effects of acute hyperglycemia on the expression of MPO and Cox-2 in rat kidney after ischemia-reperfusion. Methods: Adult male Sprague-Dawley rats were randomly divided into three groups: NG-Sham group, NG-I / R group, HG-Sham group, Blood reperfusion (HG-I / R) group 4 (n = 8). The right kidney was excised and the renal ischemia-reperfusion model was established by clamping the contralateral kidney peduncle for 45 min and reperfusion for 24 h. The rat acute hyperglycemic model was prepared by intraperitoneal injection of 25% glucose 3 g · kg-1 45 min before surgery. Renal MPO was detected by colorimetric assay. Cox-2 mRNA was detected by RT-PCR. The protein expression of Cox-2 was detected by Western blotting. Results: Compared with Sham group, MPO activity, Cox-2-mRNA and Cox-2 protein expression in I / R group were significantly increased (P <0.05) mRNA and Cox-2 protein expression was significantly higher than NG-I / R group (P <0.05). CONCLUSION: Acute hyperglycemia can increase the MPO activity, increase the expression of Cox-2, and aggravate the inflammatory injury induced by renal ischemia and reperfusion in rat kidney after ischemia-reperfusion.
