本研究对63例心、脑血管疾病患者进行了服药后3h及(或)1周的临床观察,发现穿心莲(APN)对1及5min血小板聚集率均有明显抑制作用(P<0.001),且随用药时间延长(1周后)疗效更显著。通过20名志愿者服药前后血浆及血小板5-羟色胺(5-HT)测定证明APN能显著抑制血小板释放5-HT(P<0.01)。透射电镜观察到无论体外加药还是服药后APN均能明显抑制ADP诱聚所致的血小板管导系统扩张及颗粒释放。其抗血小板聚集作用可能与激活血小板膜系统的腺苷环化酶,提高血小板内cAMP有关。
In this study, 63 patients with cardiac and cerebrovascular diseases were observed at 3h and/or 1 week after drug administration. It was found that Andrographis paniculata (APN) significantly inhibited platelet aggregation at 1 and 5 minutes (P<0.001). , And with the prolonged use of time (1 week later) the effect is more significant. The plasma and platelet serotonin (5-HT) levels before and after administration in 20 volunteers demonstrated that APN could significantly inhibit platelet 5-HT release (P<0.01). Transmission electron microscopy showed that APN inhibited platelet tube conduction system expansion and granule release caused by ADP priming in vitro and in vivo. Its anti-platelet aggregation may be related to activation of adenylyl cyclase in the platelet membrane system and increase of cAMP in platelets.
