目的 检测人骨肉瘤细胞高转移亚系SOSP M1 在裸小鼠体内传代过程中的生物学稳定性并鉴定其组织学特性。 方法 利用原位移植将细胞系在 33只裸鼠体内连续传代 ,组织块培养收集各代肺转移灶的肿瘤细胞 ,观察各代肿瘤细胞致瘤率、转移率、形态结构、骨形成蛋白、波形蛋白、肌动蛋白、神经元特异性烯醇化酶的表达情况以及遗传学方面的变化。 结果 各代肿瘤细胞致瘤率均为 10 0 % ,体内增殖稳定 ,肺转移率在 80 %以上。其显微及超微结构形态、抗原表达、染色体数目及结构变化均符合人骨肉瘤的特征。 结论 该细胞亚系在裸鼠体内传代生物学特性相对稳定 ,是人骨肉瘤实验研究的良好模型。
Objective To detect the biological stability of human osteosarcoma cell high metastatic subunit line SOSP M1 during passage in nude mice and identify its histological characteristics. Methods Cell lines were serially passaged in 33 nude mice using orthotopic transplantation. Tumor cells in each generation of lung metastases were collected and cultured to observe the tumorigenesis rate, metastasis rate, morphological structure, bone morphogenetic protein and waveform of each generation of tumor cells. Protein, actin, neuron-specific enolase expression, and genetic changes. Results The tumorigenesis rate of all generations of tumor cells was 100%. The proliferation in vivo was stable and the lung metastasis rate was above 80%. The microscopic and ultrastructural morphology, antigen expression, chromosome number, and structural changes are all consistent with the characteristics of human osteosarcoma. Conclusion This cell line has relatively stable biological characteristics in nude mice and it is a good model for human osteosarcoma experimental study.
