目的:分析2-脱氧-D-葡萄糖对血管瘤生长抑制和血管内皮生长因子(vascular endothelial growth factor,VEGF)的影响效果。方法:采用组织块移植法建立40只BALB/c血管瘤裸鼠,利用随机数字表法分为观察组和对照组,每组各20只。观察组腹腔注射2-脱氧-D-葡萄糖(500 mg/kg),对照组腹腔注射0.9%氯化钠注射液(500 mg/kg),两组均肌注苯甲酸雌二醇注射液0.1 mg。7 W后对比两组BALB/c血管瘤裸鼠移植瘤瘤体体积、阳性细胞指数、VEGF及其受体表达水平、病理组织切片、CD31免疫组化染色结果。结果:0 W时,对照组和观察组移植瘤瘤体体积差异无统计学意义(n P>0.05);1、3、5、7 W后,观察组移植瘤瘤体体积均小于对照组,差异有统计学意义[1 W:(0.12±0.01)cmn 3比(0.15±0.02)cmn 3,3 W:(0.10±0.02)cmn 3比(0.18±0.01)cmn 3,5 W:(0.08±0.01)cmn 3比(0.23±0.01)cmn 3,7 W:(0.05±0.01)cmn 3比(0.30±0.01)cmn 3,n t值分别为1.70、1.72、1.73、1.75,n P值均<0.05];对照组与观察组在不同时间段(0、1、3、5、7 W)移植瘤瘤体体积差异均有统计学意义(n F值分别为2.11和2.00,n P值均0.05);在1、3、5、7 W时,观察组阳性细胞计数均小于对照组,组间差异有统计学意义[1 W:(28.85±1.08)%比(37.40±1.10)%,3 W:(26.10±1.10)%比(42.55±1.15)%,5 W:(23.30±1.12)%比(48.14±1.16)%,7 W:(19.65±1.05)%比(52.00±1.00)%,n t值分别为1.85、1.93、2.11、2.21,n P值均<0.05];对照组和观察组在不同时间段(0、1、3、5、7 W)阳性细胞计数差异均有统计学意义(n F值分别为2.45、2.24,n P值均0.05);在1、3、5、7 W时,观察组VEGF及其受体表达水平均小于对照组,组间差异有统计学意义[VEGF:1 W:(17.34±1.08)μg/mL比(27.33±1.07)μg/mL,3 W:(15.10±1.12)μg/mL比(35.10±1.10)μg/mL,5 W:(12.87±1.11)μg/mL比(44.82±1.08)μg/mL,7 W:(9.89±1.21)μg/mL比(55.25±1.05)μg/mL,n t值分别为1.85、2.02、2.21、2.40,n P值均<0.05;VEGF受体:1 W:(20.28±1.02)μg/mL比(27.72±1.08)μg/mL,3 W:(17.46±1.04)μg/mL比(33.69±1.11)μg/mL,5 W:(13.61±1.05)μg/mL比(40.34±1.16)μg/mL,7 W:(8.87±1.03)μg/mL比(48.87±1.13)μg/mL ,n t值分别为1.82、1.93、2.17、2.33,n P值均<0.05];对照组和观察组的VEGF及其受体表达水平在不同时间段(0、1、3、5、7 W)差异均有统计学意义(VEGF:n F值分别为2.58、2.24,n P值均<0.05;VEGF受体:n F值分别为2.57、2.24,n P值均 0.05); At 1, 3, 5 and 7 W, the volume of transplanted tumor in the observation group was smaller than that in the control group, and the difference was statistically significant [1 W: (0.12±0.01) cm n 3 vs (0.15±0.02) cmn 3, 3 W: (0.10±0.02) cmn 3 vs (0.18±0.01) cmn 3, 5 W: (0.08±0.01) cmn 3 vs (0.23±0.01) cmn 3, 7 W: (0.05±0.01) cmn 3 vs (0.30±0.01) cmn 3,t values were 1.70, 1.72, 1.73 and 1.75 respectively, all n P values <0.05]; There were significant differences in the volume of transplanted tumor between the control group and the observation group at different time points (0, 1, 3, 5 and 7 W) ( n F values were 2.11 and 2.00 respectively, both n P values 0.05); at 1, 3, 5 and 7 W, the number of positive cells in the observation group was lower than that in the control group [1 W: (28.85±1.08)% vs (37.40±1.10)%, 3 W: (26.10±1.10)% vs (42.55±1.15)%, 5 W: (23.30±1.12)% vs (48.14±1.16)%, 7 W: (19.65±1.05)% vs (52.00±1.00)%, n t values were 1.85, 1.93, 2.11 and 2.21 respectively, all n P values <0.05]; The difference of positive cell count between the control group and the observation group at different time periods (0, 1, 3, 5, 7 W) was statistically significant ( n F values were 2.45 and 2.24 respectively, both n P values 0.05); At 1, 3, 5 and 7 W, the VEGF and VEGF receptor expression levels in the observation group were lower than those in the control group [VEGF: 1 W: (17.34±1.08) μg/mL vs (27.33±1.07) μg/mL, 3 W: (15.10±1.12)μg/mL vs (35.10±1.10) μg/mL, 5 W: (12.87±1.11) μg/mL vs (44.82±1.08) μg/mL, 7 W: (9.89±1.21) μg/mL vs (55.25±1.05) μg/mL,n t values were 1.85, 2.02, 2.21 and 2.40 respectively, all n P values <0.05; VEGF receptor: 1 W: (20.28±1.02) μg/mL vs (27.72±1.08) μg/mL, 3 W: (17.46±1.04) μg/mL vs (33.69±1.11) μg/mL, 5 W: (13.61±1.05) μg/mL vs (40.34±1.16) μg/mL, 7 W: (8.87±1.03) μg/mL vs (48.87±1.13) μg/mL, n t values were 1.82, 1.93, 2.17 and 2.33 respectively, all n P values <0.05]; VEGF and VEGF receptor expression levels in control group and observation group were significantly different in different time periods (0, 1, 3, 5, 7 W) (VEGF: n F values were 2.58 and 2.24 respectively, both n P values <0.05; VEGF receptor: n F values were 2.57 and 2.24 respectively, both n P values <0.05). The results of pathological sections and CD31 immunohistochemical staining showed that the number of red blood cells and positive cells in the observation group were less than those in the control group.n Conclusion:2-deoxy-D-glucose can inhibit the growth of hemangiomas, down-regulate the expression of VEGF and its receptors, and reduce the formation of new blood vessels.
