目的:探讨幽门结扎型胃溃疡寒、热证模型的形成机制及寒、热性方剂对胃溃疡寒、热证模型的基本作用机理并进一步探讨方证相应的客观规律。方法:采用寒、热因素结合幽门结扎模型建立大鼠胃溃疡寒、热证模型,采用酶联免疫法检测受试大鼠血清cAMP、cGMP的含量。结果:各模型组大鼠血清cAMP的含量与空白对照组比较明显降低,cGMP的含量显著升高(P<0.05或P<0.01)。热性模型血清cGMP含量显著高于寒性模型。大黄黄连泻心汤可显著升高热性模型组大鼠血清cAMP的含量,降低cGMP的含量(P<0.05或P<0.01);理中丸可显著升高寒性溃疡组大鼠血清cAMP的含量,降低cGMP的含量(P<0.05或P<0.01)。结论:通过“方证相应”理论验证了“病证结合”的胃溃疡寒、热证模型复制成功,大黄黄连泻心汤对热性胃溃疡、理中丸对寒性胃溃疡的治疗作用,可通过促进cAMP含量的释放、降低cGMP的含量而实现。
OBJECTIVE: To investigate the formation mechanism of cold and heat syndrome of pylorus ligation-type gastric ulcer and the basic mechanism of action of cold and heat prescriptions on cold and heat syndrome model of gastric ulcer and to further explore the objective law corresponding to the syndrome differentiation. Methods: Cold and heat factors combined with pyloric ligation model were used to establish the rat model of gastric ulcer caused by cold and heat. Enzyme-linked immunosorbent assay was used to detect the contents of cAMP and cGMP in serum of rats. Results: Compared with the blank control group, the content of cAMP in the model group was significantly decreased and the content of cGMP was significantly increased (P <0.05 or P <0.01). The content of cGMP in the heat model serum was significantly higher than that in the cold model. Rhubarb Huanglixinxin Decoction can significantly increase the heat of the rat model of cAMP content and reduce the content of cGMP (P <0.05 or P <0.01); RZ can significantly raise the level of serum cAMP in rats with cold ulcer, decreased cGMP content (P <0.05 or P <0.01). Conclusions: The model of gastric ulcer caused by cold and heat syndrome was successfully verified by the theory of “Correspondence of square prescription”, and the effect of rhubarb Huanglianxixin decoction on hyperthermic gastric ulcer, Therapeutic effect can be achieved by promoting the release of cAMP content and reducing cGMP content.
