三磷酸腺苷结合盒转运蛋白(ATP-binding cassette,ABC)广泛分布于各种生物体中,是最大的膜蛋白家族之一。在昆虫中,ABC转运子不仅在分子转运过程中有重要的功能,同时在杀虫剂抗性、代谢和发育中都起到重要的作用。克隆和鉴定了褐飞虱(Nilaparvata lugens Stal,BPH)ABCG基因(Nl ABCG),结果表明Nl ABCG基因全长1 222 bp,含有一个长度为789 bp的开放阅读框,编码263个氨基酸。Nl ABCG蛋白与内华达古白蚁(Zootermopsis nevadensis)ABCG的同源性最高,达到69%。利用q RTPCR检测Nl ABCG基因在褐飞虱不同发育阶段和不同组织中的表达,发现Nl ABCG转录本在褐飞虱的所有发育阶段都有表达,在1龄若虫中表达量最低,随着生长发育表达量逐渐升高。Nl ABCG在褐飞虱5龄若虫的中肠中表达量最高。Nl ABCG基因的序列特征和表达谱非常保守,为其功能的分析提供了信息。
ATP-binding cassette (ABC) is widely distributed in various organisms and is one of the largest family of membrane proteins. In insects, ABC transporters not only play an important role in molecular transport, but also play an important role in insecticide resistance, metabolism and development. The ABCG gene (Nl ABCG) of Nilaparvata lugens Stal (BPH) was cloned and identified. The results showed that the Nl ABCG gene was 1 222 bp in length and contained an open reading frame of 789 bp, encoding 263 amino acids. The Nl ABCG protein had the highest homology with ABCG of Zootermopsis nevadensis, reaching 69%. The qRTPCR was used to detect the expression of Nl ABCG gene at different developmental stages and different tissues of N. lugens. Nl ABCG transcripts were found at all developmental stages of N. lugens, and were the lowest in the first instar nymphs. With the development and development, Rise. Nl ABCG showed the highest expression in the midgut of the 5th instar nymphs of BPH.
