目的:评价咪哒普利对大鼠心肌梗死后8周心室重构和MMPs的影响。方法:冠状动脉左前降支结扎后24h,手术组大鼠被随机分至对照组(C组,n=10)、咪哒普利组(I组,n=10)。另设冠状动脉未结扎的大鼠为假手术组(SH组,n=10)。治疗后8周行超声心动图检查,心室称质量,明胶酶谱法测定左心室心肌组织MMPs的活性。结果:与SH组比较,手术组大鼠的相对左室质量(LVRW)、相对右室质量(RVRW)和左室舒张末内径(LVEDD)显著升高;与C组比较,I组可显著降低LVRW、RVRW和LVEDD。与SH组比较,C组的MMP-2活性显著升高;与C组比较,I组的MMP-2活性显著降低。结论:咪哒普利可抑制大鼠心肌梗死后8周MMP-2的活性,改善心室重构。
AIM: To evaluate the effect of imidapril on ventricular remodeling and MMPs 8 weeks after myocardial infarction in rats. Methods: The rats in the operation group were randomly divided into control group (C group, n = 10) and imidapril group (group I, n = 10) after ligation of left anterior descending coronary artery. The other rats without coronary artery ligation were sham operation group (SH group, n = 10). Echocardiography was performed at 8 weeks after treatment, and ventricular mass was measured. Gelatin zymography was used to determine the activity of MMPs in left ventricular myocardium. Results: The LVRW, RVRW and LVEDD in the operation group were significantly higher than those in the SH group. Compared with the C group, the I group was significantly decreased LVRW, RVRW and LVEDD. Compared with SH group, the activity of MMP-2 in group C was significantly increased. Compared with group C, the activity of MMP-2 in group I was significantly decreased. CONCLUSION: Imidapril inhibits MMP-2 activity and improves ventricular remodeling at 8 weeks after myocardial infarction in rats.
