目的利用实时荧光定量PCR(real time polymerase chain reaction,real-time PCR)技术测定IFN-γmRNA的表达水平,并研究其在肾移植患者术后免疫抑制治疗疗效评价中的应用。方法取肾移植后患者外周血分离淋巴细胞,提取总RNA,使用定量PCR测定干扰素(interferon,IFN)-γmRNA表达水平。结合治疗药物监测结果和临床检验结果分析。结果 IFN-γ对免疫抑制方案、肾功能、移植后时间和年龄不同的肾移植患者使用免疫抑制剂的不同药效有较好的区分。结论 IFN-γ可作为肾移植免疫抑制剂的药效学指标,为个体化给药提供依据。
Objective To determine the expression of IFN-γmRNA using real-time PCR (real-time PCR) technique and to evaluate the therapeutic effect of immunosuppressive therapy in renal transplant recipients. Methods Lymphocytes were isolated from the peripheral blood of patients after kidney transplantation and the total RNA was extracted. The expression of interferon (IFN) -γmRNA was measured by quantitative PCR. Combined with the treatment of drug monitoring results and clinical test results analysis. Results IFN-γ had a better distinction between immunosuppressive agents, different efficacy of immunosuppressive agents in immunosuppressive regimen, renal function, time and age after transplantation. Conclusion IFN-γ can be used as a pharmacodynamic indicator of immunosuppressive agents for renal transplantation, providing a basis for individualized administration.
