目的研究NF-κB及PARR-γ在Barrett食管及食管腺癌中转录水平的表达,探讨Barrett食管(BE)及食管腺癌的发病机制。方法采用real-time PCR法检测正常食管上皮、Barrett食管黏膜及食管腺癌标本中NF-κB mRNA及PARR-γmRNA表达情况。结果NF-κB mRNA在BE及食管腺癌中表达均增强,其表达率平均为正常鳞状上皮的1.2058倍(P<0.01)和1.7212倍(P<0.01);PARR-γmRNA在BE及食管腺癌中表达明显增强,其表达率分别为正常鳞状上皮的5.9130倍(P<0.01)和2.0314(P<0.01)。结论NF-κB及PARR-γ转录水平明显增高说明两者的表达上调在BE及食管腺癌的发生发展中可能起着重要作用。
Objective To investigate the expression of NF-κB and PARR-γ in Barrett’s esophagus and esophageal adenocarcinoma and to explore the pathogenesis of Barrett’s esophagus (BE) and esophageal adenocarcinoma. Methods The expression of NF-κB mRNA and PARR-γmRNA in normal esophageal mucosa, esophageal mucosa of Barrett and esophageal adenocarcinoma were detected by real-time PCR. Results The expressions of NF-κB mRNA in BE and esophageal adenocarcinomas were significantly increased, the expression rate of NF-κB mRNA was 1.2058 folds (P <0.01) and 1.7212 folds (P <0.01) higher than that of normal squamous epithelium Compared with normal squamous epithelium, the positive expression rate was 5.9130 times (P <0.01) and 2.0314 (P <0.01) respectively. Conclusions The transcriptional level of NF-κB and PARR-γ is significantly increased, indicating that upregulation of both may play an important role in the development of BE and esophageal adenocarcinoma.
