目的探讨芩丹胶囊(QC)含药血清对动脉血管外膜成纤维细胞(AF)的作用机制。方法采用组织贴块法体外原代培养大鼠胸主动脉成纤维细胞,MTT法观察大[750mg/(kg·d)]、小[150mg/(kg·d)]剂量的QC含药血清对转化生长因子β1(TGF-β1)诱导的大鼠胸主动脉成纤维细胞的增殖的影响;实时定量PCR及western-blot分别检测α-平滑肌肌动蛋白(α-SMactin)和Smad3的基因及蛋白表达情况。结果大、小剂量QC含药血清呈剂量依赖性抑制TGF-β1诱导的AF的增殖(P<0.05),且用药组α-SMactin和Smad3基因及蛋白的表达较单纯刺激组显著降低(P<0.05)。结论QC含药血清通过干扰TGF-β1/Smad信号转导通路抑制AF增殖和分化,从而改善和逆转动脉血管外膜重构。
Objective To investigate the mechanism of action of Qindan capsule (QC) containing serum on arterial adventitial fibroblasts (AF). Methods Thoracic aorta fibroblasts were primary cultured in vitro by tissue patch method. The concentrations of QC-containing serum in large [750 mg / (kg · d)] and small [150 mg / (kg · d)] were observed by MTT assay TGF-β1 induced thoracic aorta fibroblasts proliferation; Real-time quantitative PCR and western-blot were used to detect the α-SMactin and Smad3 gene and protein Express the situation. Results Large and small doses of QC-containing serum could inhibit the proliferation of AF induced by TGF-β1 in a dose-dependent manner (P <0.05), and the expressions of α-SMactin and Smad3 gene and protein were significantly decreased (P < 0.05). Conclusion QC-containing serum can improve and reverse the adventitia remodeling by interfering with TGF-β1 / Smad signal transduction pathway to inhibit the proliferation and differentiation of AF.
