为了揭示层粘连蛋白(laminin,Ln)信号传递作用与转移性肺腺癌增殖的关系。体外以外源性Ln(exogenousLn,exLn),抗Ln抗体(antibodyofLn,Ln-Ab)以及PMA[phorbol-12-myristate-13-acetate,蛋白激酶C(PKC)激活剂]作用于小鼠肺腺癌内源性Ln(endogenousLn,enLn)高表达的高转移克隆化正系(C6)及其低表达的低转移克隆化正系(C7)进行比较研究。结果:exLn能使C6细胞比C7细胞的增殖力在已是较高水平的基础上进一步提高,并与PMA的共同作用显示了增殖效应的协同。相反,Ln-Ab能使C7细胞比C6细胞的增殖力获得更明显抑制;并且,仅PMA的作用却使C6细胞比C7细胞的增殖力呈现更明显的抑制,并使C6细胞的跨膜Ca2+流恒态(homeostasis)发生显著性改变。结论:层粘连蛋白信号传递作用与高、低转移性小鼠肺腺癌细胞的增殖密切相关,这可能与PKC活性及跨膜Ca2+流恒态的变化相互关联.
To reveal the relationship between laminin (Ln) signaling and the proliferation of metastatic lung adenocarcinoma. In vitro exogenous Ln (exogenous Ln, exLn), anti-Ln antibody (antibody of Ln, Ln-Ab) and PMA [phorbol-12-myristate-13-acetate, protein kinase C (PKC) activator] act on mouse lung adenocarcinoma High-endogenous Ln (enLn) high-transformation clone positive lines (C6) and their low-expression low-transition cloned positive lines (C7) were compared. RESULTS: ExLn could increase the proliferative power of C6 cells more than C7 cells on the basis of a higher level, and the co-action with PMA showed the synergistic effect of proliferation. On the contrary, Ln-Ab can make C7 cells more significantly inhibit the proliferation of C6 cells; and only the role of PMA will make C6 cells show more significant inhibition of proliferation than C7 cells, and make C6 cells transmembrane Ca2+ There was a significant change in homeostasis. Conclusion: Laminin signal transduction is closely related to the proliferation of high and low metastatic mouse lung adenocarcinoma cells, which may be related to the changes of PKC activity and the transmembrane Ca2+ current state.
