目的:探讨P-糖蛋白(P-gp)、人表皮生长因子受体2(HER-2)及microRNA-296的表达与食管癌放射抗拒的相关性。方法:将人食管癌细胞Eca109分为对照组和处理组,处理组通过X射线反复照射(累计放射剂量60 Gy),采用MTT法检测2组细胞的增殖抑制率。免疫细胞化学法测定P-gp和HER-2的表达。Northern blotting法测定microRNA-296的表达。结果:处理组细胞较对照组显示出明显的放射抗性,增殖抑制率明显低于对照组细胞。免疫细胞化学测定P-gp和HER-2在处理组中较对照组表达显著增加。2组细胞中mi-croRNA-296表达无显著差异。结论:P-gp、HER-2可能与食管癌放射抗拒有关;暂不能说明microRNA-296与食管癌细胞Eca109放射抗拒具有相关性。
Objective: To investigate the relationship between P-glycoprotein (P-gp), human epidermal growth factor receptor 2 (HER-2) and microRNA-296 expression and radioresistance in esophageal cancer. Methods: The human esophageal carcinoma cell line Eca109 was divided into control group and treatment group. The treatment group was repeatedly irradiated by X-ray (the cumulative dose was 60 Gy). The proliferation inhibition rate of the two groups was detected by MTT assay. Immunocytochemistry was used to determine the expression of P-gp and HER-2. Northern blotting assay microRNA-296 expression. Results: Compared with the control group, the treated group showed obvious radioresistance and the inhibition rate of proliferation was significantly lower than that of the control group. Immunocytochemistry showed that P-gp and HER-2 were significantly increased in the treatment group compared with the control group. There was no significant difference in mi-croRNA-296 expression between the two groups. Conclusion: P-gp and HER-2 may be related to radioresistance of esophageal cancer. However, we can not conclude that microRNA-296 is associated with radioresistance of esophageal cancer cell Eca109.
