为了研究他莫西芬(TAM),酯化雌二醇(E_2)以及生理浓度的17β-E_2在体外抑制内皮细胞介导的低密度脂蛋白(LDL)氧化的作用,对其作用进行了比较。采用胶原酶消化的方法分离得到人脐静脉内皮细胞(HUVEC),并保存于特定的培养基及培养条件下。先将HUVEC分别与生理浓度的17β-E_2溶液、E_217酯、TAM以及α-维生素E(阳性对照)、乙醇溶液(阴性对照)共孵育48小时,再取空腹状态下血脂正常人的肝素经血浆分离LDL,分别与各种溶液共孵育的HUVEC共同培养(每1 ml无血清Ham F-10培养基中包含100μgLDL)8、16、
In order to investigate the effects of tamoxifen (TAM), estrified estradiol (E2), and physiological concentrations of 17β-E_2 on endothelial cell-mediated inhibition of low density lipoprotein (LDL) oxidation in vitro, their effects were compared . Human umbilical vein endothelial cells (HUVECs) were isolated by collagenase digestion and stored in specific medium and culture conditions. HUVECs were incubated with 17β-E 2 solution, E 2 17 ester, TAM and α-tocopherol (positive control) and ethanol solution (negative control) for 48 hours at physiological concentrations respectively.
